摘要
为了研究乳酸杆菌LA-5对雏鸡肠道黏蛋白MUC2含量变化的影响,应用实时荧光定量PCR技术和间接酶联免疫吸附试验对饲喂乳酸杆菌雏鸡回肠、盲肠、直肠MUC2mRNA及相应肠液MUC2蛋白含量进行检测。结果发现:饲喂乳酸杆菌雏鸡肠道MUC2mRNA表达及肠道MUC2含量在饲喂后1~18d均不同程度高于对照雏鸡,其中直肠MUC2mRNA表达水平在饲喂后1—7d极显著高于对照组(P〈0.01);回肠肠液MUC2含量于饲喂后1~4d显著高于对照组(P〈0.05)。表明乳酸杆菌对雏鸡肠道组织黏蛋白MUC2基因表达和分泌均有影响,能促进肠道黏液分泌,提高雏鸡肠道黏膜免疫功能。
To investigate the effect of Lactobacillus LA-5 on the content of mucin-MUC2 in intestinal tract of chicks,expression of MUC2 mRNA in ileum,cecum and rectum and the content of MUC2 in corresponding intestinal fluid were detected by quantitative real-time PCR and indirect enzyme-linked immunosorbent assay. The results showed that after fed LA-5 for 1 to 18 days,the content of MUC2 in intestinal fluid from three kinds of intestinal tissues and MUC2 mRNA expression were higher than the control group in different extents. The expression of MUC2 mRNA in rectum was very significantly enhanced compared with the control group after fed LA-5 for 1 to 7 days (P〈0.01). Meantime, the content of MUC2 in intestinal fluid from ileum tissue was also significantly enhanced compared with the control group after fed LA-5 for 1 to 4 days (P〈0.05). Above all,the expression of MUC2mRNA and secretion of MUC2 could be both up-regulated by LA-5,whieh could promote the secretion of intestinal mucus and then enhance intestinal mucosal immunity of chicks.
