摘要
目的观察4周跑台运动对慢性应激致去卵巢大鼠抑郁、焦虑行为及空间学习记忆能力和前额叶皮质(Prefrontal Cortex,PFC)单胺类神经递质水平及脑源性神经营养因子(Brain-Derived Neurotrophic Factor,BDNF)表达的影响。方法 40只SD大鼠随机分为假手术组、去卵巢组、去卵巢应激组及去卵巢应激运动组,除了假手术组大鼠,其余3组大鼠采用卵巢摘除建立大鼠围绝经期模型,再通过慢性不可预知性应激(chronic unpredictable stress,CUMS)建立抑郁模型,同时,应激运动组大鼠进行4周跑台运动。运动及应激结束后通过旷场实验、高架迷宫实验、明暗箱探索实验及水迷宫实验等方法测试大鼠行为学指标变化,采用高效液相-电化学检测法检测前额叶皮质去甲肾上腺素(norepinephrine,NE)、多巴胺(dopamine,DA)及5-羟色胺(5-hydroxy trptamine,5-HT)的含量,用免疫组织化学结合图像半定量方法对前额叶皮质BDNF神经元的数量、面积进行测量和分析。结果卵巢摘除及慢性不可预知应激增强了大鼠的抑郁行为及焦虑行为,学习记忆能力下降,行为学表现为旷场试验中大鼠穿越格数及直立次数均显著减少,高架迷宫实验中OE%、OT%及(开放臂+闭合臂)均显著下降,明暗箱实验中大鼠明箱时间缩短,水迷宫实验中学习记忆能力下降;血清皮质醇(cortisol,Cort)水平增加,前额叶皮质单胺类神经递质DA、NE、5-HT含量均显著下降,BDNF表达减弱。而长期跑台运动可以显著改善去卵巢抑郁模型大鼠的抑郁样行为及增强学习记忆能力,血清Cort水平下降,前额叶皮质单胺类神经递质DA、NE、5-HT含量增加,BDNF表达增强。结论跑台运动可改善去卵巢抑郁模型大鼠的抑郁及焦虑行为,增强大鼠学习记忆能力,机理可能与长期跑台运动降低去卵巢抑郁大鼠Cort水平、拮抗HPA轴功能亢进、上调前额叶皮质BDNF的表达、有效调整中枢神经递质紊乱有关。
Objetive To explore the effects of the 4 - week treadmill exercise on the depressive behavior and learning and memory, monoamine levels and BDNF expression within PFC in the ovariectomized rats subjected to Chronic Unpredictable Stress (CUMS). Methods 40 female SD rats were randomly allocated to 4 groups, which included sham - operated rats ( SHAM), ovariotomy rats (OVX) , stressed OVX rats ( OVX + S) and stressed OVX rats treated with exercise ( OVX + S + E). All rats except SHAM group rats established perimenopausal model by ovarieetomy and depression model by chronic unpredictable stress, then OVX + S + E group rats receiced 4 - week treadmill exercise at the same time. The behavioral changes were conducted by open field test ( OFT), elevated plus - maze test (EPM) , light - dark (LD) exploration and Morris water maze( MWM), then the monoamine levels ( NE, DA and 5 - HT) were examined by high performance liquid - electrochemical detection and BDNF expression in PFC was detected and analyzed by using ABC immunohistochemical tech- nique and semi quantitative. Results We found that the rats subjected to ovariectomy and CUMS increased depressive and anxiety behavior and impaired learning and memory, as measured crossing numbers and rearing numbers significantly de- creased in OFT; OE% ,OT% and (OE + CE) numbers significantly reduced in EPM; light box time shortened in LD and impair learning and memory in MWM ; the serum cort increased, the NE, DA and 5 - HT levels significantly decreased and BDNF expression weakened in PFC. Results showed that treadmill exercise may significantly improve depressive behavior and enhance learning and memory of the ovariectomized depressive rats; the cort concentration significantly decreased, the NE, DA and 5 - HT levels significantly increased and BDNF expression enhanced in PFC in the OVX + S group compared with the OVX + S + E group.
