摘要
【目的】分生孢子在树木腐烂病菌传播过程中起着极其重要的作用,是进行病原菌研究的重要实验材料,获得形态和萌发率一致的分生孢子尤为重要。【方法】本研究采用不同方法获得病菌的分生孢子,置于不同环境条件下,测定其萌发率筛选最优获取方法和萌发条件。【结果】田间采集发病组织获得分生孢子较为容易但易被污染;接种发病枝条和30%PBA培养基15 d左右均可获得形态一致和萌发率较为一致的分生孢子。【结论】病菌分生孢子萌发最适温度为25℃、相对湿度100%、pH为5.0-6.0、黑暗条件并需要一定的营养物质。
[Objective]Conidia plays an important role in the spread of tree canker pathogens,and it is an major experimental material in studying rot fungi. Therefore,It is significant to gain condia with almost the same morphology and germination rate. [Methods]Pathogen conidia obtained with different methods,was placed in distinct circumstances to measure germination rate in order to select optimal acquisition method and germination conditions. [Results]It is easier to obtain conidia with which the incidence tissues directly collected from field,but it is easier polluted; Condia with consistent germination rate and morphology can be attained both in case of inoculated in disease branches and 30 % PBA culture for about 15 d. [Conclusion]The conidia germination optimum temperature is25 ℃,relative humidity 100 %,and pH 5. 0-6. 0,under the dark condition and certain nutrients.
