摘要
目的 观察乙酰肝素酶(HPSE)抑制剂对Hela细胞导致的裸鼠皮下移植瘤模型的生长及血管生成的抑制作用.方法 将18只Hela细胞裸鼠皮下移植瘤模型随机分为空白对照组(A组)、紫杉醇联合卡铂作用组(B组)及HPSE抑制剂作用组(C组),每组6只,空白对照用生理盐水代替药物,采用腹腔灌注方式给药,观察13 d后处死裸鼠,比较各组移植瘤体积变化,行肿瘤组织CD34免疫组织化学染色,观察肿瘤模型微血管密度(MVD).结果 第13天测量A、B、C3组肿瘤体积分别为(1 218 ±182)、(573±85)、(627±101) mm3,A组肿瘤生长速度快于B组和C组,差异有统计学意义(P<0.05);A、B、C3组MVD值分别为49.5 ±5.8、21.8±4.7、24.2 ±4.9,B组和C组MVD值小于A组,差异有统计学意义(P<0.05).结论 HPSE抑制剂可以有效抑制Hela细胞裸鼠皮下移植瘤的生长及血管的生成.
Objective To investigate the inhibitory effects of heparanase (HPSE) inhibitor on growth and angiogenesis of subcutaneous xenotransplanted tumor model of Hela cells in nude mice.Methods Eighteen subcutaneous xenotransplanted tumor models were randomly allocated into blank control group (group A),paclitaxel plus carboplatin group (group B) and HPSE inhibitor group (group C) with 6 in each group.Blank control group was disposed with 0.9% NaCl solution instead of medicine.All groups were dosed through intraperitoneal injection and executed 13 days after injection.Tumor volume was measured and microvessel density (MVD) of tumor model was detected through immunohistochemical staining of CD34.Results At 13th day after the palpable tumors formed,we found the average tumor volume in groups A,B and C was (1218 ± 182),(573 ± 85) and (627 ± 101) mm3 respectively.The average tumor volume in group A was significantly larger than in groups B and C (P <0.05 for both).MVD in groups A,B and C was 49.5 ± 5.8,21.8 ± 4.7 and 24.2 ± 4.9 respectively.MVD in groups B and C was significantly less than that in group A (P < 0.05).Conclusion HPSE inhibitor can effectively suppress the growth and angiogenesis of subcutaneous xenotransplanted tumor models of Hela cells in nude mice.
