摘要
目的研究培养基中添加不同浓度的萘乙酸(NAA)对诱导铁皮石斛组培苗的影响。方法以铁皮石斛种子为材料,在4种培养基(I^IV)中诱导出原球茎,再以预实验优选出的1/2MS+活性炭1.0g/L+蔗糖30g/L+琼脂7.0g/L为基本培养基,配制成0~5.0 mg/L的6种不同NAA浓度的培养基(V^X)进行原球茎增殖分化和继代培养,筛选促进组培苗根茎叶生长发育最佳的NAA浓度和培养基配方。结果种子在IV号培养基中的萌发率最高,达94.2%以上;在NAA添加量为4.0 mg/L的IX号培养基中,原球茎分化后根长、叶长、叶宽、株高、茎粗、单株鲜重分别为1.516 cm、0.943 cm、0.365cm、2.991 cm、0.264 cm、0.102 g,各指标均显著优于其它NAA浓度的培养基。另外,在IX号培养基中添加100 g/L土豆和80 g/L香蕉能够显著地促进生根壮苗阶段组培苗根茎叶的生长发育,各指标可增加1~3倍。结论培养基1/2MS+NAA0.2 mg/L+蔗糖30.0 g/L+琼脂7.0 g/L最适宜铁皮石斛种子萌发,当NAA浓度为4.0 mg/L时更能促进组培苗根茎叶的生长发育。
Objective This research was conducted to study the effects on the D.candidum plantlets at various concentrations of naphthylacetic acid(NAA) supplied to the basic medium.Methods At first,the D.candidum seeds were sowed in four media(I ~ IV) for germinating and obtaining protocorms.Then the protocorms were transplanted to the pre-screened medium of 1 /2MS + activated carbon 1.0 g / L + sucrose 30.0 g / L + agar7.0 g / L with 6 different NAA concentrations(V ~ X),from 0 mg / L to 5.0 mg / L,to achieve the subculture,differentiation and proliferation of protocorms.After 5 months of growing,from each medium bottles,11 plantlets were randomly selected for getting the traits data measured for the growth of roots,stems and leaves,which was conducted for screening optimal NAA concentration and medium formulations for D.candidum plantlets during tissue culturing.Results The medium IV got the highest germination rate of 94.2%.The plantlets planted in IX medium got the maximum values in traits of root length,leaf length,width,height,stem diameter,plant fresh weight with exact data of 1.516 cm,0.943 cm,0.365 cm,2.991 cm,0.264 cm and 0.102 g,significantly better than the other mediums with others NAA concentrations.In addition,these trait data could be enhanced one or three times when the IX medium were added muds of 100 g / L potatoes or 80 g / Lbananas.Conclusion The medium of 1 /2MS + NAA 0.2 mg / L + sucrose 30 g / L + agar 7.0 g / L is the most suitable for seed germination of Dendrobium D.candidum.The 4.0 mg / L is the optimum NAA concentration promoting the seedling growths on leaf,root and stem in tissue culture of D.candidum.
