兔眼角膜碱烧伤后骨桥蛋白与角膜新生血管的相关性研究 被引量:1

Study on the correlation between OPN and corneal neovascularization after corneal alkali burn

摘要 目的建立兔眼角膜碱烧伤新生血管动物模型,并探讨房水中骨桥蛋白(OPN)与角膜新生血管(CNV)的相关性,为研究CNV的形成机制提供实验依据。方法选取内蒙古医科大学动物实验中心2014年4月64只12月龄的新西兰大耳兔进行实验,体重2.0~2.5 kg,雌雄不限,裂隙灯显微镜检查实验兔无眼疾,常规饲养。采用数字表法将兔随机分成A、B、C、D、E、F、G、H组,每组8只兔。其中A组为对照组,其余7组为实验组。碱烧伤制作CNV动物模型,不同时间点测量各组CNV长度并抽取兔双眼房水,计算CNV面积并检测房水中OPN的浓度。CNV面积和OPN浓度以均数±标准差(x珋±s)的形式表示,并采用单因素方差分析的方法进行组间比较,当比较结果有统计学意义时,进一步采用SNK法进行组间两两比较。结果 A组兔角膜透明无血管,角膜缘血管网呈环形并不侵入角膜。角膜碱烧伤后3 d对应的B组炎症反应较重,结膜充血水肿,角膜水肿,角膜碱烧伤斑明显,CNV面积为(23.88±0.89)mm2;5 d后对应的C组CNV面积为(27.59±0.73)mm2;7 d后对应的D组角巩膜缘处可见CNV芽呈毛刷状长入角膜,角巩膜缘血管明显扩张,CNV面积为(34.46±0.72)mm2;11 d后对应的E组CNV生长旺盛,均匀致密,部分侵入烧伤斑,CNV面积为(38.54±0.56)mm2;14 d对应的F组CNV致密呈网状,血管粗大多数长至角膜中央区,CNV面积为(41.95±0.34)mm2;21 d对应的G组新生血管较多较粗,但数量有所减少,CNV面积(41.93±0.33)mm2;28 d对应的H组CNV变得稀疏,分支也减少,CNV面积为(39.81±0.89)mm2,各组间CNV面积比较,差异有统计学意义(F=72.41,P〈0.05)。角膜碱烧伤后A组对应时间点房水OPN浓度平均为(0.47±0.10)μg/L,角膜碱烧伤3 d后对应的B组房水OPN浓度为(1.75±0.07)μg/L,变化较为明显,与对照组比较,差异有统计学意义(P〈0.05);7 d后所对应的C组OPN浓度为(5.51±1.18)μg/L,此时OPN� Objective To establish the relationship of osteopontin (OPN) with corneal neovascularization (CNV) in a rabbit model of alkali corneal burns. Methods Sixty-four New Zealand big- eared rabbits, weighing 2.0-2.5 kg and aged 12 months,were randomized to 8 groups ( i. e. ,A, B, C, D, E, F,G and H, n = 8). Animals in group A were left untreated and served as controls, while those in the remaining 7 groups were subjected to corneal burns with 2 mol/L sodium hydroxide. At different time points after corneal burns, the length and area of the newly formed small vessels in the injured cornea were assessed and OPN concentrations in the aqueous humor collected from the eye were measured by ELISA. Results In the control group, the cornea was transparent without either new blood vessel formation or invasion of the limbal vascular network in the ring into the corneal area ( CNV -- 0 mm2 ). In animals with experimental alkali corneal burns, inflammation aggravated gradually between post-burn day 3 and day 14 and conjunctival hyperemia and edema and corneal edema were significantly more evident as compared with control animals (P 〈 0.05 ) ;from day 14 to day 28 conjunctival hyperemia and edema, corneal edema, and corneal alkali burns became stable. Low OPN concentration (0.47±0.10 μg/L) was detected in the corneal aqueous humor in control animals. In animals with experimental corneal burns, OPN concentration was gradually increasing from day 3 to day 14 (P 〈0.05) but was gradually decreasing from day 14 d to day 28 (P 〈0.05). Conclusion Corneal angiogenesis is closely correlated with OPN concentrations in the aqueous humor of the eye, suggesting an important role for OPN in. corneal neovascularization after burn injury.
