摘要
【目的】研究一株石油降粘菌株BT-003的生长特性及对石油的降粘效果。【方法】采用比色法、东秀珠细菌鉴定方法和单因素试验,研究菌株BT-003生长特性,考察菌液不同接种量、不同反应时间对石油降粘效果的影响。【结果】菌株BT-003培养4 h开始进入对数生长期,10 h后即为稳定期,可以利用石油为唯一碳源进行生长,35℃下具有最大菌体密度;5%菌液接种量与石油作用8-14 h即可达到很好的降粘效果,石油黏度由30 000 m Pa·s降低到150 m Pa·s。【结论】菌株BT-003在石油中生长繁殖快,用量少,耗时短,适用于黏度较大油井的三次开采。
【Objective】To understand the growth characteristic of strain BT- 003 and affecting factors of the bacteria which were used in viscosity reduction of oil.【Method】The growth characteristic of the strain BT- 003 and the influence of different inoculation and time on the viscosity reduction of oil were studied by using colorimetric method,bacteria identification method and the single factor test.【Result】The results showed that the strain BT- 003 arrived in logarithmic phase after 4h and stable phase after 10 h. The strain BT- 003 as the only carbon source could be cultured on oil. The most density of bacteria was reached at the temperature35℃. The consequence of the single factor test showed 5% inoculation,8- 14 h effecting with oil produced good effect and the oil viscosity was achieved from 30 000 m Pa·s to 150 m Pa·s.【Conclusion】The strain BT- 003 grew quickly in the oil and achieved good viscosity reduction effect by using less dosage and time,which was proved to be very suitable for the oil production.
