摘要
为了解MHC-B单倍型在武定鸡的分布情况与促进抗马立克病(MD)品系的选育,采用对血细胞DNA进行微卫星PCR扩增及测序的方法,对20个武定鸡试验鸡群进行分型,并对雏鸡进行马立克病毒(MDV)京-1株攻毒试验。结果发现,鸡群中B11单倍型出现频率最高(约为37%),同时发现了B21单倍型纯合子(约为4%);攻毒试验表明,B5/B5、B15/B15、B11/B11、B14/B14和B21/B21对MD易感性依次降低。本研究初步鉴定了武定鸡群中各MHC-B单倍型分布情况,并建立了筛选武定鸡抗MD品系的方法。
To understand the MHC-B haplotype distribution in Wuding chicken and promote the breeding of Marek's disease ( MD )resistant lines, we applied PCR mierosatellite and DNA sequencing to blood cell samples.Twenty test groups of Wuding gen- otypcd and chicks were ehallenged with J-1 Marek's disease virus (MDV). It was found that B11 haplotype had the highest fre- quency (approximately 37% )and there was B21 haplotype homozygote (approximately 4% ) ; virus challenge test showed that the susceptibility to MD in B5/B5, B 15/B 15, B 11/B 11 , B 14/B 14 and B21/B21 gradually decreased. This study preliminarily identified MHC-B haplotypes distribution in Wuding flock and established methods for screening anti-MD Wuding chicken lines.
