摘要
目的检测Rab27A与Rab27B在3种人肝癌细胞系及1种永生化肝细胞系中的表达情况,初步研究Rab27A与Rab27B表达对肝癌细胞生物学特性的影响。方法采用RT-PCR及Western blot技术,检测Rab27A与Rab27B mRNA在3种人肝癌癌细胞Hep G2、Huh-7、Li-7及1种永生化肝细胞系HL-7702中的表达。结果Rab27A与Rab27B在肝癌细胞系及原发性肝癌组织中存在差异性表达。Rab27A 在3种人肝癌细胞系中表达明显强于1种人永生化肝细胞系;Rab27B mRNA在3种人肝癌及1种人永生化肝细胞系中均明显表达但无显著差异。结论 Rab27A与Rab27B与原发性肝癌密切相关,可以为临床采取合理诊断措施提供有效的参考依据。
Objective To investigate the expression of Rab27A and Rab27B in 3 human HCC lines and the immortal-ized hepatic cell line and its effect on biology characteristics of the cells. Methods The mRNA expression of Rab27A and Rab27B was detected by RT-PCR,and Western-blot in 3 human HCC lines of HepG2,Huh-7,Li-7 and the immor-talized hepatic cell line of HL-7702. Results Rab27A and Rab27B were differentially expressed in cell lines and pri-mary HCC tumors.The expression of Rab27A in 3 human HCC line was significantly better than that expressed in the immortalized hepatic cell line;the mRNA of Rab27B in 3 human HCC lines and the immortalized hepatic cell line were significantly expression but there was no significantly difference. Conclusion Rab27A and Rab27B expression are closely correlated HCC,it can provide the clinical reasonable diagnostic measures as valuable effective reference basis.
