摘要
采用硫酸二乙酯(diethyl sulfate,DES)诱变方法对火木层孔菌菌株进行诱变处理,研究了0.5%诱变浓度下DES诱变时间与火木层孔菌致死率的关系。对筛选的优势菌株菌丝生长速度、液体发酵菌丝体生物量、菌丝形态、色素类次级代谢产物分泌以及次级代谢产物抗氧化活性方面的差异进行研究。研究表明,硫酸二乙酯诱变可改变菌丝形态但对菌丝体生长并未产生明显影响,且硫酸二乙酯诱变筛选可改善药用火木层孔菌活性成分,提高对缺氧/糖PC12神经细胞的抗氧化活性。
The diethyl sulfate (DES) mutagenesis was chosen for the mutagenic treatment to Phellinus igniarius, and the relation- ship of mutagenesis time and death rate was investigated with 0. 5% DES. The differences of mycelial growth speed, liquid fermentation mycelia biomass, morphology and pigment c/asses of secondary metabolites production speed and antioxidant activities of metabolite products were discussed. The study displayed that DES mutagenesis could change mycelial morphology without obvious effect on myceli- um growth, and the DES mutagenesis improved antioxidant activities of the active ingredients of P. igniarius and had more antioxidant activity of hypoxia/sugar PCI2 nerve cells than that of P. igniarius.
