摘要
基于腺病毒的靶向基因治疗是肿瘤生物治疗方式之一.为了更好的了解腺病毒靶向药物在裸鼠实体瘤中的运作效果,本研究中利用重组腺病毒报告载体Ad LacZ分别转染人肿瘤细胞系和裸鼠实体瘤,结合X-gal染色和组织切片的方法观察腺病毒载体在实体瘤原位转染后的运作模式,为实体瘤实验提供更多的经验借鉴和实验基础.
Adenovirus vector mediated gene therapy offers a new choice for the cancer, but is not sufficient to work when it is used alone. In order to understand the operation process of adenovirus vector in vivo, we investigated the efficiency infection of such as repli- cation-defective adenovirus Ad LacZ in cancer cells and solid tumor of nude mice, the results showed that the infection effect of Ad LacZ associated with the density of cells. The research provided the experimental basis for the application of replication-defective adenovirus in cancer therapy.
