摘要
目的:建立同时测定痤疮酊中氯霉素和水杨酸含量的方法。方法:采用高效液相色谱法。色谱柱为Shim-pack CLCODS,流动相为甲醇-水(50∶50,V/V),流速为0.8 ml/min,检测波长为278 nm,柱温为35℃,进样量为10μl。结果:氯霉素和水杨酸质量浓度分别在2.0~10.0μg/ml(r=0.999 2)、5.0~25.0μg/ml(r=0.999 1)范围内与各自峰面积呈良好的线性关系;精密度、稳定性、重复性试验的RSD〈1%;氯霉素和水杨酸的平均加样回收率分别为99.77%、99.29%,RSD分别为0.35%、0.48%(n=9)。结论:该方法操作简便、快速,结果准确,专属性好、精密度高,可用于痤疮酊的质量控制。
OBJECTIVE:To establish a method of microbial limit test for Ofloxacin gel. METHODS:Test sample solutions were prepared by Mn SO4,Ca Cl2 and Mg Cl2 gelout with different concentrations,the numbers of bacteria and control bacteria were detected by membrane filtration method,and the numbers of molds and yeasts were determined by conventional method. RESULTS:The conventional method showed the recoveries of Candida albicaus and Aspergillus niger were more than 70% after not treated by gel breaker;the membrane filtration method showed the recoveries of Escherichia coli,Staphylococcus aurus and Bacilus subtilis were more than 70% after treated by 5% Mn SO4,3% Ca Cl2 and 1% Mg Cl2;the membrane filtration method could detect the control bacteria after treated by gel breaker. CONCLUSIONS:Gel breaker with appropriate concentrations can effectively eliminate the antibacterial activity of Ofloxacin gel. The established method is suitable for its microbial limit test.
