摘要
目的通过观察当归芍药散水煎剂(DSS)对于垂体后叶素(Pit)引发的心肌缺血的作用效果,评定当归芍药散对于心肌缺血的治疗价值。方法选取昆明种小鼠80只,随机分为空白组、模型对照组、DSS干预组(给予DSS水煎剂灌胃)以及阳性对照组(给予复方丹参滴丸灌胃),连续干预7 d后,在末次药后1 h,除空白组小鼠外,其余小鼠应用Pit建立心肌缺血模型,分别于造模前、造模后10 min、15 min以及20 min时进行心电图检查观察小鼠心率变化,应用免疫组化法检测血清一氧化氮(NO)水平、内皮型一氧化氮合酶(e NOS)活性,同时测定心肌组织中超氧化物歧化酶(SOD)活性以及丙二醛(MDA)含量。结果 1建立模型后,与空白处相比,各组小鼠的心率均显著下降(P<0.05),但是与模型对照组相比,DSS干预组和阳性对照组小鼠的心率改善显著(P<0.05)。2DSS干预组血清NO、心肌e NOS水平显著高于模型对照组(P<0.05)。3造模后,模型小鼠的SOD活性显著低于空白组,MDA含量显著高于空白组,DSS干预组和阳性对照组小鼠SOD活性显著高于模型对照组(P<0.05),而且MDA含量显著低于模型对照组(P<0.05)。结论当归芍药散能够有效保护心肌细胞,减轻Pit所致心肌缺血的损伤程度。
Objective Through observing the effect of Danggui Shaoyao San Decoction( abbreviated as DSS)for pituitrin-induced myo-cardial ischemic injury,to assess the value of Danggui Shaoyao San for the treatment of myocardial ischemia. Methods 80 Kunming mice were selected into this study. They were randomly divided into blank group,model control group,DSS treatment group( treated with DSS decoction ga-vage)and positive control group( treated with Compound Danshen Dropping Pill gavage). After 7 d continuous intervention,1 h after the last ad-ministration of medicine,myocardial ischemia model were established by Pit in the other mice apart from blank control group mice. ECG examina-tion were given in 10 min,15 min and 20 min before and after modeling and mice heart rate changes were observed recorded. The immunohisto-chemical method was used to detect the nitric oxide( NO)level of serum,endothelial nitric oxide synthase( eNOS)activity. At the same time, the myocardial tissue superoxide dismutase( abbreviated SOD)activity and malondialdehyde( MDA)content were detected. Results ①After the establishment of the model,the heart rate in mice were significantly decreased in the other groupscompared with the blank group( P 〈0. 05),but heart rate was significantly improved in DSS intervention group and positive control group( P 〈0. 05)compared with the model control group. ②DSS group serum NO level,myocardial eNOS level was significantly higher than that in model control group( P 〈0. 05). ③In DSS group,myo-cardial SOD activity was significantly higher than the model group( P 〈0. 05). In DSS group,myocardial MDA content was significantly better than the model group( P 〈0. 05). Conclusion Danggui Shaoyao San Decoction can effectively protect myocardial cell and reduce the damage degree of myocardial ischemia induced by Pit.
