摘要
选取5个西番莲株系的一年生枝条进行不同梯度的低温胁迫处理,观察受冻情况并测定质膜相对透性、抗氧化酶活性、渗透调节物质等指标,分析其抗寒能力。结果表明:相对电导率随处理温度的降低逐渐升高,0℃以下呈对数上升趋势;不同株系间MDA含量变化趋势不同,差异显著(P<0.05);SOD活性随处理温度降低呈"先降后升"或"降-升-降"的变化趋势;平塘1号、版纳4号的CAT活性较低且变幅不大,其他株系的CAT活性随处理温度降低呈逐渐下降趋势;不同株系间POD活性不同且处理间的变化趋势不同,抗寒株系其活性较高;可溶性蛋白含量随处理温度降低迅速增加,0℃含量达最大值,温度继续降低则出现下降趋势;可溶性糖与脯氨酸含量随处理温度降低呈现逐渐上升趋势。运用隶属函数法进行综合评价,得出抗寒性强弱顺序为:平塘1号>版纳9号>版纳4号>紫香1号>版纳10号。
This study selected one year-old branches from different Passiflora edulis cultivars to be tested for chilling stress in response to different temperature treatments. The cold tolerance was evaluated by determining the relative membrane permeability, antioxidant enzyme activity, concentration of osmotic solutes and other in- dicators. The results showed that the relative conductivity gradually increased while temperature dropped; the relative conductivity increased in a logarithmical manner when the temperature was below 0 ℃. Different trends of changes in MDA content among different cultivars were detected (P〈0.05). SOD activity first de- creased then increased as the temperature kept dropping, or showed a trend of "decrease-increase-decrease". CAT activity in Pingtang No. 1 and Banna No.4 was low and stayed unchanged. In contract, CAT activity de- creased in other cultivars while temperature decreased. POD activity in different cultivars showed distinct trends of changes in response to different treatments; higher activity was detected in cultivars with higher cold tolerance. Soluble protein content increased rapidly as temperature decreased; the content reached maximum at 0 ℃. However, the content of soluble proteins started to decrease when the temperature dropped below 0 ℃. The content of soluble sugar and proline showed a gradual increase with dropping temperature. Comprehensive evaluation using the membership function indicated that the ranking of cold tolerance of the tested Passiflora edulis cultivars was as Pintanz No. 1 〉Banna No.9〉Banna No.4〉Zixiang No. 1 〉Banna No. 10.
