摘要
目的探讨微小RNA-205(miRNA-205)在非小细胞肺癌(NSCLC)组织和血浆中的表达和临床意义。方法用实时定量PCR法测定31例NSCLC患者(试验组)及10例肺良性病变患者(对照组)支气管镜活检组织和血浆中miRNA-205的表达量,分析其与各临床病理资料之间关系,评价miRNA-205在NSCLC的诊断价值。结果试验组活检组织及血浆miRNA-205表达量均高于对照组(P<0.05)。miRNA-205在鳞癌组织中的表达高于腺癌(P<0.05)。miRNA-205的表达量与NSCLC患者临床病理特征无明显相关(rs=0.255,P=0.113)。ROC曲线分析显示,取界值0.932时,组织miRNA-205表达诊断NSCLC的灵敏度和特异度分别为81.3%和83.3%;取临界值0.610时,血浆miRNA-205表达诊断NSCLC的灵敏度和特异度分别为81.3%和84.2%。结论 miRNA-205在NSCLC组织和血浆均有较高的表达,可能会成为诊断NSCLC,尤其是鳞癌的参考指标。
Objective To investigate the expression and significance of microRNA（miRNA）- 205 in non-small cell lung cancer（NSCLC） tissues and plasma. Methods With qRT-PCR, the expressions of miRNA-205 in NSCLC tissues obtained from bronchoscopy and in plasma were detected in group A（31 patients with NSCLC） and group B（10 patients with lung binign lesions）. The correlation between miRNA-205 expressions and clinieopathologic features of NSCLC was analyzed. The value of rniRNA-205 expressions in the diagnosis of NSCLC was evaluated as well. Results The expressions of miRNA-205 were significantly higher in group A than those in group B（P〈0. 05）. The miRNA-205 expression was significantly higher in squarnous cell carcinoma （SCC） than that in adenocarcinoma（AD） （P〈0. 05）. Plasma miRNA-205 expression was not significantly correlated with clinicopathologic features of NSCLC（rs=0. 255, P=0. 113）. ROC curve analysis showed that taking 0. 932 as a cutoff value, the sensitivity and specificity for miRNA-205 expression in tissues to diagnose NSCLC were 81.3% and 83.3%, which for miRNA-205 expression in plasma were 81.3% and 84. 2% when 0. 610 was taken as a cutoff value. Conclusion The miRNA-205 is highly expressed in NSCLC tissues and plasma of NSCLC patients. The level of miRNA-205 expression could be considered as a potential diagnostic marker for NSCLC, especially for SCC.
