摘要
目的:探究咖啡及其主要成分胡芦巴碱对2型糖尿病大鼠的胰岛素敏感性、脂代谢、炎症反应以及脂联素水平等的影响。方法:以高脂高糖饲料喂养联合腹腔注射小剂量链脉佐菌素建立2型糖尿病大鼠模型,所有大鼠随机分成四组:正常对照组、模型组、咖啡组和胡芦巴碱组。测定空腹血糖(FBG)、胰岛素(FINS)、胰岛素敏感指数(ISI)、总胆固醇(TC)、甘油三醋(TG)、高密度脂蛋白胆固醇(HDL)、肿瘤坏死因子α(TNF-α)、白介素-6(IL-6)和脂联素(APN)等水平的变化。结果:咖啡和胡芦巴碱均可降低2型糖尿病大鼠的血糖和胰岛素水平,并显著提高胰岛素敏感性(P<0.05);胡芦巴碱可使TC和TG水平显著下降(P<0.05),HDL水平显著升高(P<0.05),而咖啡只能使TC水平降低(P<0.05);咖啡组IL-6水平显著下降(P<0.05),APN水平显著升高(P<0.05),而胡芦巴碱还能显著降低TNF-α水平。结论:咖啡和胡芦巴碱均可有效提高胰岛素敏感性,改善脂代谢紊乱,减轻炎症反应以及促进APN分泌。
Objective:To investigate the effects of coffee and trigonelline on insulin sensitivity,lipid metabolism, inflammation reaction and the level of APN in type 2 diabetes mellitus rats.Method:Type 2 diabetic rats were rendered by feeding high energy diet together with injection of streptozotocin (STZ).The rats was randomly divided into the control group,the model group,the coffee group and the trigonelline group.The changes of glucose,insulin sensitivity,lipids,TNF-α,IL-6 and adiponectin (APN) were observed.Result:Both coffee and trigonelline could reduce glucose and insulin levels and raise insulin sensitivity(P〈0.05).Trigonelline could lower TC and TG levels and increase HDL level(P〈0.05),but coffee could only reduce TC levels (P〈0.05).In coffee group,the level of IL-6 was down and the level of APN was up (P〈0.05),more than that, trigonelline could lower the level of TNF-α(P〈0.05).Conclusion:Coffee and trigonelline could effectively increase insulin sensitivity,improve lipid metabolic disorder,alleviate the inflammation reaction,and promote the secretion of APN.
