摘要
目的:探讨液氮快速冻融法水蛭提取物(HE)对人肝癌Hep G2细胞的化疗药物敏感性影响及诱导凋亡的机制。方法:以5-氟尿嘧啶(5-FU)、阿霉素(ADM)为对照,MTT法检测HE与5-FU或AMD联合用药对人肝癌Hep G2细胞的增殖抑制作用,流式细胞术测定细胞凋亡;采用逆转录聚合酶联反应(RT-PCR)法、Western blot法检测Hep G2细胞经HE处理后多药耐药基(MDR1)、Bax、Caspase3基因的m RNA及蛋白表达变化。结果:HE可增强5-FU、ADM对Hep G2细胞的增殖抑制作用及诱导细胞凋亡(P<0.01);可显著下调Hep G2细胞MDR1(P<0.01)和上调Caspase3 m RNA及蛋白表达(P<0.01)。结论:HE可体外诱导Hep G2细胞凋亡,并提高Hep G2细胞对化疗药物5-FU的敏感性。其分子机制与下调MDR1表达,上调Caspase3表达有关。
Objective: To study the effects and mechanism of himdo extract (HE) on the sensitivity of chemotherapeutic drugs in human hepatoma HepG2 cells. Methods: HepG2 cells were treated in vitro by HE combined with 5-FU or ADM as the control groups. The growth suppression and apoptosis were examined by MTT and flow cytometry assay respectively. Expressions of mRNA and protein of MDR1, Bax and Caspase3 were detected by RT-PCR and Western blot methods in HepG2 cells treated by HE. Results: HE enhanced the suppression of HepG2 cells proliferation caused by treatment of 5-FU or ADM (P〈0.01). The mRNA and protein expression of MDR1 was down-regulated (P〈0.01) and Caspase3 was up-regulated significantly (P〈0.01). Conclusion: HE can induce apoptosis and enhance the chemotherapeutic sensitivity of 5-FU and ADM in HepG2 cells through the mechanism of down-regulating the expression of MDR1 and up-regulating Caspase3.
