白虎汤对内毒素致热家兔的解热作用及其机制研究 被引量:21

摘要 目的探讨白虎汤对内毒素发热家兔的解热作用及其机制。方法将32只新西兰大白兔随机分为正常组、模型组、白虎汤低、高剂量组(10,20 g/kg),造模前,空白、模型2组分别灌胃等量生理盐水,白虎汤低、高剂量组分别灌胃不同剂量药物,1次/d,连续5 d。第6天,除正常组外,其余3组分别通过耳缘静脉注入大肠杆菌内毒素方法复制发热家兔模型,观察各组动物症状及体温变化;采用双抗体夹心ELISA法检测血清细胞因子IL-1β、IL-6、肿瘤坏死因子TNF-α水平;采用流式细胞仪测定T淋巴细胞亚群CD4+、CD8+水平。结果与正常组相比,模型组家兔体温,TNF-α、IL-1β、IL-6、ET和CD8+水平显著上升(P<0.01),CD4+水平与CD4+/CD8+显著下降(P<0.01);与模型组相比,白虎汤高、低剂量组家兔体温、TNF-α、IL-1β、IL-6、ET、CD8+水平明显下降(P<0.05或P<0.01),CD4+水平与CD4+/CD8+显著上升(P<0.05或P<0.01)。白虎汤高、低剂量均对内毒素性发热家兔有显著的解热效应(P<0.05或P<0.01),均可明显抑制发热家兔TNF-α、IL-1β、IL-6、ET、CD8+水平升高(P<0.05或P<0.01),促进CD4+水平与CD4+/CD8+显著上升(P<0.05或P<0.01)。结论白虎汤对内毒素性发热家兔有解热的作用,机制可能是通过抑制发热家兔致热性细胞因子的释放,调节机体免疫功能而发挥其解热的作用。 Objective To explore the antipyretic effect and mechanism of Baihu decoction for fever rabbits caused by endotoxin. Methods The 32 New Zealand rabbits were randomly divided into normal group,model group,Baihu decoction low-dose group,high-dose group( 10,20 g / kg). Before modeling,blank group and model group,two groups were given amount of normal saline respectively,Baihu decoction low-dose group,high-dose group were given doses of drugs respectively,once a day,and lasted five days,The sixth day,except the normal group,the other 3 groups were injected through ear vein Escherichia Coli Endotoxin replication fever in rabbits model,to observe the change of symptoms and body temperature of each animal; Using double antibody sandwich Ell SA method to test the levels of serum IL-1β,IL-6,tumor necrosis factor TNF-a. Using flow cytometry to test the levels of Tlymphocyte subsets and CD4+,CD8+. Results Compared with normal group,the model group significantly increased body temperature in rabbits,TNF-α,IL-1β,IL-6 and CD8+levels( P 〈0. 01),CD4+level and CD4+/ CD8+decreased significantly( P 〈0. 01);Compared with the model group,Baihu decoction high dose group significantly decreased in low dose group rabbits body temperature,TNF-α,IL-1β,IL-6 the level of CD8+( P 〈0. 05,P 〈0. 01),increased CD4+levels and CD4+/CD8+( P 〈0. 05 or P 〈0. 01). Baihu tang Low-dose group,high dose group both had antipyretic effect significantly for fever rabbits caused by endotoxin( P 〈0. 05 or P 〈0. 01),can inhibit the fever in rabbits TNF-α,IL-1β,IL-6,CD8+levels( P 〈0. 05 or P 〈0. 01),promote the significant increase of the CD4+level and CD4+/ CD8+( P 〈0. 05 or P 〈0. 01). Conclusion The Baihu decoction has antipyretic effect on rabbits with endotoxin fever,mechanism may be the inhibition on fever in rabbits induced heat release of cytokines,regulate immune function and its antipyretic role.
