摘要
采用PCR-SSCP和测序技术检测促黄体素β亚基基因5′调控区和3个外显子序列在小尾寒羊、多赛特羊、萨福克羊和乌珠穆沁羊中的单核苷酸多态性,分析其多态性与小尾寒羊产羔数的相关性。结果表明:引物P2、P4和P5扩增片段均存在多态性。引物P2对应的突变[A(-424)G、T(-271)C]以及引物P4对应的突变[G(-6)A]均会引起调控区结合因子的显著变化。对于P2座位,AA型小尾寒羊产羔数显著高于其余基因型,其余基因型之间小尾寒羊产羔数差异均不显著;对于P4和P5座位,各基因型之间小尾寒羊产羔数差异均不显著。由此推断该基因5′调控区序列A(-424)G和T(-271)C的单碱基突变可能是提高小尾寒羊产羔数潜在的有效标记。
To elucidate the polymorphism of ovine luteinizing hormone β subunit (LH β) gene and its relationship with litter size of Small Tail Han sheep, single nucleotide polymorphisms (SNPs) of 5' regulatory region and all three exons (exons 1, 2 and 3) of LH,8 gene were detected in Small Tail Han sheep, Dorset sheep, Suffolk sheep and Ujumqin sheep by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing technology. The products amplified by primers P2, P4 and P5 displayed polymorphism. Software analysis results revealed that mutations detected by P2 [A(-424)G, T(-271)C] and mutation detected by P4 [G(- 6)A] caused significant changes between binding factors with 5'regulatory region of LH,8 gene. For primer P2, the litter size of genotype AA was higher (P〈 0.05) than those with other genotypes, but there were no significant differences between other genotypes (P〉0.05) in Small Tail Han sheep. For primer P4 and primer P5, differences of litter size between genotypes were not significant (P〉 0.05) in Small Tail Han sheep. These results preliminarily indicated that A(-424)G and T(-271)C polymorphic sites of 5' regulatory region of LH β gene were potential markers for improving litter size in Small Tail Han sheep.
