摘要
目的探讨芪卫颗粒对糖尿病肾病(diabetic nephropathy,DN)大鼠肾组织足细胞裂孔隔膜蛋白CD2相关蛋白(CD2-associated protein,CD2AP)表达的影响。方法采用数字表法随机选取10只大鼠为正常组,其余大鼠采用腹腔注射大剂量链脲佐菌素(streptozotocin,STZ)(60 mg/kg)诱导DN大鼠模型,将成模大鼠采用数字表法随机分为模型组、西药组以及芪卫颗粒组,连续给药,动态监测大鼠血糖、体质量、24 h尿微量白蛋白;24周后将大鼠处死,采用免疫组织化学法及蛋白印迹法(Western blotting)检测肾组织足细胞CD2AP表达情况。结果与正常组相比,模型组呈高血糖、低体质量状态(P<0.05),24 h尿微量白蛋白浓度显著升高,足细胞CD2AP表达显著下降(P<0.05);与模型组相比,各给药组体质量、血糖无明显变化,24 h尿微量白蛋白浓度明显下降,CD2AP表达明显上调(P<0.05)。结论芪卫颗粒能够上调CD2AP在肾组织的表达,降低DN大鼠尿微量白蛋白排泄,延缓DN的发生、发展。
Objective To investigate the effects of Qiwei granules on the expression of CD2-associated protein(CD2AP) in podocytes of rats with diabetic nephropathy ( DN ) . Methods Ten normal rats were chosen randomly as the normal group, and the others were abdominally administrated with a single injection of streptozotocin(60 mg/kg) to induce diabetic nephropathy rat models. The model rats were separated into 3 groups randomly:the model group, the western medicine group and Qiwei granules group, and successive administration for 24 weeks. Fasting blood glucose( FBG) , body weight( BW) , 24 h urinary albumin excretion( UAER) were examined every 4 weeks. After 24 weeks, all the rats were sacrificed, and CD2AP in podocyte was detected by immunohistochemistry and Western blotting. Results As compared with normal control group, the BW in model group are significantly decreased(P〉0. 05), meanwhile the FBG and 24 hUAER are significantly increased(P0. 05), 24 h UAER is decreased(P〈0. 05), and the expression of CD2AP is significantly increased(P〈0. 05). Conclusion 24 h UAER is decreased in Qiwei granules group. The protective effects of Qiwei granules group on kidney are achieved by up-regulating the expression of CD2AP.
