摘要
[目的]探讨小麦Ta NRX基因与抗旱的关系及其调控机制。[方法]根据晋麦Ta NRX基因序列,设计一对特异引物,利用RTPCR法从"平凉45号"冬小麦的c DNA中扩增出一个DNA片段,并克隆到p MD19-T载体,测序后比对分析。[结果]该片段具有完整的开放阅读框,长度为1 734 bp,编码577个氨基酸,与Genbank中晋麦的核酸序列具有100%的同源性,氨基酸序列中包含2个氧化还原二硫化物活性中心WCGPC和WCPPC,属于TRX家族,同时生物信息学分析发现该基因编码不稳定、非分泌、脂溶性蛋白。实时荧光定量PCR检测显示,该基因在"平凉45号"冬小麦各器官中均有表达,但具有不同的表达模式。[结论]为进一步深入研究Ta NRX基因的功能及与其他基因之间的相互关系提供参考。
[ Objective ] To study the relationship between TaNRX gene and drought resistance of wheat. [ Method ] According to Jin wheat TaN- RX gene sequence, a pair of specific primers was designed and a DNA fragment was amplified from the Pingliang 45 winter wheat using RT-PCR method, then it was cloned into pMD19-T vector, comparison and analysis was conducted after sequencing. [Result] The fragment has a com- plete open reading frame, 1 734 bp in length, encoding a protein of 577 amino acids. It has 100% homology with the Genbank nucleic acid se- quence in Jin wheat, contains WCGPC and WCPPC. The cloning gene belonged to the TRX family, and the code is not stable, non secretion abd fat soluble protein. The gene was all expressed in different organs of Pingliang No. 45 of winter wheat, but with a different expression pattern by the real-time fluorescence quantitative PCR. [ Conclusion] This study provides a reference to further study the relationship or function between TaNRX gene and other genes.
