摘要
目的:研究电针足三里对对乙酰氨基酚(Acetaminophen,APAP)诱发大鼠肝损伤炎症细胞因子水平的影响,探讨其对药源性肝损伤的保护作用。方法:SD大鼠按随机数字表法分为NC组、APAP1组和Acup1组、APAP2组和Acup2组,各组大鼠连续给药或针刺14 d后,检测大鼠血清和肝脏组织匀浆液白细胞介素6(IL-6)、白细胞介素8(IL-8)和肿瘤坏死因子α(TNF-α)的含量。结果:与APAP1组和APAP2组比较,Acup1组和Acup2组大鼠血清谷草转氨酶(AST)和谷丙转氨酶(ALT)活性水平均显著性升高。与APAP1组和APAP2组比较,Acup1组和Acup2组大鼠血清和肝脏匀浆组织液中IL-6、IL-8和TNF-α含量水平均显著升高。结论:电针足三里上调APAP诱导大鼠肝损伤血清和肝脏组织炎症细胞因子的反应和表达水平,可能会对APAP诱导的肝脏损伤具有一定的促进作用。
Objective : To investigate the effects of acupuncture at Zusanli aeupoint on inflammatory eytokines changes in rats with given multiple doses of paracetamol, and the protective effect of acupuncture on hepatic injury. Methods: SD rats were divided into five groups, the NC group, the APAP1group and the Acupl group,the APAP2 group and the Acup2 group. Each group was given paracetamol or saline once a day for 14 continuous days, and the serum levels of alanine transaminase(ALT) and aspartate transaminase (AST)of the rats were measured, and the levels of Interleukin 6 (IL-6), Interleukin 8 (IL-8), tumor necrosis factor-α (TNF-α) in serum and liver tissue homogenate were determined by euzymelinked immunosorbent assay(ELISA) respectively. Results: Compared with groups of APAPl andAPAP2, the levels of serum AST and ALT were significantly increased in groups of Acup1 and Acup2. And compared with groups of APAP~ andAPAP2, the levels of IL-6,IL-8 and TNF-α in the serum and liver were significantly increased in groups of Acup1 and Acup2. Conclusion : The results suggest that electroacupuncturing Zusanli acupoints can increase levels of inflamatory IL-6, IL-8 and TNF-α in the serum and liver, and may promote liver damage induced by paracetamol.
