摘要 目的探讨microRNA-214(miR-214)在女性乳腺癌患者血清中的表达水平及其潜在的临床意义。方法提取35例乳腺癌术前患者、25例乳腺癌术后患者、25例良性乳腺肿瘤患者及25例健康女性血清中的总RNA,用实时荧光定量PCR检测血清miR-214的表达水平,并分析其与乳腺癌患者临床病理参数之间的关系。结果乳腺癌术前组血清miR-214的表达水平明显高于术后组(Z=-3.40,P〈0.01)、良性乳腺肿瘤组(Z=-2.33,P〈0.05)及健康对照组(Z=-4.72,P〈0.01)。miR-214在乳腺癌术前组中区别于健康个体中的ROC曲线下面积(AUCROC)为0.847(95%CI:0.744~0.950),当cut off值为4.42时,敏感性与特异性分别为97.1%和96.0%。在乳腺癌术前组中miR-214的表达水平与淋巴结的转移(Z=-2.05,P〈0.05)及年龄(Z=2.85,P〈0.01)有关。结论 miR-214在乳腺癌患者血清中的表达水平升高,可能作为评估乳腺癌淋巴结转移的一项辅助指标。 Objective To investigate the expression level of serum mieroRNA-214 (miR-214) in female breast cancer patients and its potential clinical significance. Methods The total RNAs were extracted from serum samples of 35 patients with preoperative breast cancer, 25 with postoperative breast cancer, 25 with benign tumors and 25 healthy women, respectively. Then, the expression level of serum miR-214 was detected by real-time PCR, and the correlations of miR-214 expression level with pathological parameters of breast cancer were evaluated. Results The expression levels of miR-214 in patients with preoperative breast cancer were significantly higher than those in patients with postoperative breast cancer ( Z = - 3.40, P 〈 0.01 ), patients with benign tumor ( Z = - 2.33, P 〈 0.05 ), and healthy women (Z = -4.72, P 〈 0.01 ), respectively. Serum miR-214 could be used to distinguish preoperative breast cancer pa- tients from healthy women, and the area under curve (AUC) of ROC analysis was 0. 847 (95% CI : 0. 744-0. 950). When the cut off was 4.42, the sensitivity and specificity were 97.1% and 96.0% , respectively. The expression levels of serum miR-214 in preopera- tive breast cancer patients were correlated with lymphatic metastasis ( Z = - 2.05, P 〈 0.05 ) and age ( Z = 2.85, P 〈 0.01 ) of breast cancer patients. Conclusion The increased serum miR-214 levels in breast cancer patients may be used as an auxiliary marker for e- valuating lymphatic metastasis of breast cancer.
