摘要
目的:优选经方配方颗粒平胃散挥发油的提取与包合工艺并考察包合物的稳定性。方法:采用L9(34)正交试验法,以加水量、蒸馏时间、浸泡时间为考察因素,以挥发油提取量为评价指标,优选挥发油最佳提取工艺;以挥发油与β-环糊精(β-CD)的比例、包合时间、包合温度为考察因素,以挥发油利用率、包合率为综合评分指标,优选最佳包合工艺;用薄层色谱法(TLC)检测包合质量,对包合物进行光、热、湿稳定性考察。结果:最终确立挥发油的提取工艺为:10倍量水浸泡0.5h,水蒸气蒸馏6h;挥发油包合工艺为:挥发油与β-CD的比例为1∶8(V∶m),30℃下搅拌包合90min。结论:优选出的经方配方颗粒平胃散挥发油提取工艺与包合工艺稳定、合理、可行。
Objective:To optimize the preparating technology for extraction process and inclusion process of volatile oil from Ping- WeiSan and investigate the stability of the inclusion compound. Methods:The study was carried out with L9 (34 ) ortbogonal design, using three indexes as the investigation factors, namely the added water, the distillation time and soaking time, volatile oil extraction amount as evaluation indicators to choose the best extraction process; using three indexes namely the proportion of volatile oil to β-cyclodextrin(β-CD), stirring time and inclusion temperature as the investigation factors, to evaluate the volatile oil utilization efficiency,inclusion yield and inclusion oil content comprehensively,in order to select the best inclusion process. Thin layer chromatography (TLC) is used to inspect quality of inclusion in order to carry out light,heat, wet clathrate stability test. Results:Eventually the extraction process of volatile oil is:ten times the amount of water immersion 0.5 h,water vapor distillation 6 h; The volatile oil inclusion process:the volatile oil and β-CD ratio of 1 : 8 (V : m) ,and 30℃ under stirring package 90 min. Conclusion. The optimizated extraction and inclusion process of volatile oil is reasonable, feasible and available.
