柯乐猪血液基因组甲基化修饰与细胞因子水平的相关性 被引量:1

摘要 为探明柯乐猪血液免疫细胞的基因表达是否受甲基化调控,应用甲基化敏感扩增多态性技术(MSAP)和酶联免疫吸附测试技术(ELISA)对柯乐猪与大白猪血液基因组DNA的甲基化水平、血清中的细胞因子浓度进行检测,分析柯乐猪与大白猪在甲基化水平、细胞因子浓度方面的差异及其相关关系。结果表明:柯乐猪基因组总甲基化水平为32.38%,与大白猪接近(P>0.05);IL-2、IL-4、IL-6和IL-10浓度分别为(302.37±125.72)pg/mL、(88.71±32.05)pg/mL、(219.01±30.42)pg/mL和(116.95±30.24)pg/mL,其中IL-6和IL-10分别极显著、显著低于大白猪;IFN-α、IFN-γ分别为(211.09±76.08)pg/mL和(57.18±14.21)pg/mL,分别极显著、显著高于大白猪。柯乐猪基因组的全甲基化水平与IFN-γ浓度间呈弱负相关关系(ρ=-0.256)。说明,血液免疫细胞的基因表达可能受甲基化的调控,且与柯乐猪的抗病力强有关。 The methylation level of genomic DNA and cytokines concentration in serum of Kele and Yorkshine pigs were detected by MSAP and ELISA and the correlation between methylation level and cytokines concentration was analyzed to explore the effect of methylation regulation on genetic expression of blood immune cells of Kele pig.The genomic methylation level of Kele pig is 32.38% and is similar to Yorkshine pig(P〉0.05).The concentration of IL-2,IL-4,IL-6 and IL-10 is (302.37±125.72)pg/mL, (88.71±32.05)pg/mL,(219.01±30.42)pg/mL and (116.95±30.24)pg/mL respectively,of which,IL-6 and IL-10 concentration is very significantly and significantly lower than Yorkshine pig separately.The concentration of IFN-αand IFN-γ is (211.09±76.08)pg/mL and 57.18±14.21)pg/mL,very significantly and significantly higher than Yorkshine pig separately.There is a weak negative correlation between genomic methylation level and IFN-γ concentration in Kele pig (ρ= -0.256),which indicates that gene expression of blood immune cells may be regulated by methylation and be related to strong disease resistance of Kele pig.
