摘要 目的探讨甲状腺癌细胞株SW579中PP4R1对细胞增殖能力的影响。方法利用慢病毒完成甲状腺癌细胞SW579/p CDH1-Con和SW579/p CDH1-PP4R1的建株,采用Western blot和qRT-PCR技术检测PP4R1表达变化;体外用CKK-8法检测SW579细胞增殖的变化;流式细胞仪检测SW579细胞周期的变化;克隆形成实验检测两组克隆形成率的变化。结果PP4R1过表达慢病毒感染甲状腺癌SW579细胞系构建成功;在体外实验中,过表达PP4R1可抑制细胞增殖,将细胞周期阻滞于S期,从而抑制甲状腺肿瘤的生长。结论 PP4R1在甲状腺癌的发生发展中起着重要作用,其是否能作为甲状腺癌新的靶向分子仍有待进一步的研究。 Objective To investigate the effect of protein serine/threonine phosphatase 4 regulatory subunit 1(PP4R1)on pro-liferation of SW579 thyroid cancer cells .Methods Thyroid cancer cells SW579/PCDH1-Con and SW579/pCDH1-PP4R1 were built by Centivirus imfection .The expression levels of the enzyme in SW 579 cells were examined by using Western blot and qRT-PCR analy-ses.The changes of the cell proliferation were determined by using a CCK-8 assay.The changes of the cell cycle were detected by using a flow cytometry while a clone formation assay was used to observe the change in the clone formation rate .Results SW579 cell line that was infected with PP4R1 over-expression Lentivirus was successfully constructed .In in vitro experiments,over-expressed PP4R1 could inhibit the proliferation of SW 579 cells and arrest the cell cycle at S phase, thus inhibit thyroid tumor growth .Conclusion PP4R1 may play an important role in thyroid cancer development .However,further studies are needed to determine whether it can be used as a novel therapeutic target for treatment of the cancer .
