摘要
以6个具有代表性的大豆品种作为实验材料提取7S和11S,经Superdex 200凝胶柱层析纯化制得纯品7S和11S,用凯氏定氮和SDS-PAGE电泳综合分析其纯度在90%以上。采用SDS-PAGE电泳测定纯品7S和11S的亚基组分,用圆二色光谱分析纯品7S和11S的二级结构,用ANS荧光探针法测定纯品7S和11S的表面疏水性。经分析得出如下结论:大豆蛋白质的表面疏水性与α/亚基含量不相关,与α亚基含量呈正相关,与β亚基含量呈负相关;与酸性亚基含量呈负相关,与碱性亚基含量呈正相关;与α-螺旋含量呈负相关,与β-折叠含量呈负相关,与β-转角含量呈正相关,与无规则卷曲含量呈正相关。
The 7s and 11s were extracted from six representative soybean varieties which were as experimental material. The purity of 7s and 11s which purified by the Superdex 200 gel column chromatography system were more than 90%by SDS-PAGE electrophoresis and Kjeldahl. Determined by SDS-PAGE electrophoresis pure 7S and 11S subunit component,secondary structure by circular dichroism spectroscopy of pure 7S and 11S,the surface hydrophobicity of 7s and 11s was determined by the ANS fluorescence probe Method.The conclusion showed that the surface hydrophobicity of Soybean protein was not associated with the content of the subunitα;was positively correlated with the content of the subunit α;was negative correlation with the content of the subunit β;was negative correlation with the content of the Acidic subunit;was positively correlated with the content of the Basic subunit;was negative correlation with the content of theα-helix;was negative correlation with the content of the β-fold;was positively correlated with the content of the β- corner;was negative correlation with the content of the random coil.
