百脉根离体再生体系的优化 被引量:6

Optimization of Culture Conditions for Lotus japonicus Regeration in Vitro

摘要 本研究以百脉根叶片、叶柄、茎段、花柄和根组织为外植体,比较了它们在不同激素配比培养基中的愈伤组织、不定芽及不定根的分化情况,优化并建立百脉根组织培养再生体系。研究结果显示:除根外,其它组织均可通过组织培养获得再生植株;外植体类型是影响愈伤组织、不定芽及不定根诱导的主要因素,叶片外植体的诱导效果最好。优化的百脉根组织培养再生体系为:使用叶片外植体,愈伤组织诱导培养基为MS+3.0 mg/L 6BA+0.1 mg/L NAA、芽分化培养基为MS+0.3 mg/L KT、生根培养基为1/2 MS+0.4 mg/L IBA。通过该体系60~75 d可获得百脉根再生植株,约为无性繁殖周期的1/3~1/2。 Legume Lotus japonicus has been widely used as a model system for genetic and molecular studies based on the classic hypocotyls transformation-regeneration procedure. Here, explants of leave, stem, petiole, flower stalk and root from Lotus japonicus were inoculated on the MS basal medium, adding different hormone for plant regeneration study. Results showed that callus and adventitious buds could be easily and efficiently induced from Lotus japonicus leave, stem, petiole and flower stalk except for roots on different culture mediums. Explants type was the major factor affecting the callus, adventitious bud and root induction. Optimum mediums for the induction of callus, adventitious bud and root were MS+3.0 mg/L 6BA+0.5 mg/L NAA, MS+0.2 mg/L KT and 1/2 MS+0.4 mg/L IBA, respectively, based on using leave explant. New regenerating lines could be obtained within 60~75 days by the optimized regeneration system.
