地奥心血康抗炎镇痛作用实验研究 被引量:2

摘要 目的研究地奥心血康(黄山药、穿龙薯蓣)的抗炎镇痛作用。方法采用福尔马林致小鼠疼痛反应模型、二甲苯致小鼠耳肿胀模型、醋酸致小鼠腹腔毛细血管通透性增高模型、棉球致小鼠肉芽肿模型和角叉菜胶致小鼠气囊炎症模型观察地奥心血康的抗炎镇痛作用;采用酶联免疫吸附法(ELISA)硝酸还原法测定气囊炎症模型小鼠气囊灌洗液中肿瘤坏死因子(TNF-α)和一氧化氮(NO)水平,采用硫代巴比妥酸(TBA)法和水溶性的四唑盐法(WST-1)测定模型小鼠血清中丙二醛(MDA)和超氧化物歧化酶(SOD)水平,分析其可能的抗炎机制。结果地奥心血康80 mg/kg、60 mg/kg能显著减少福尔马林致痛模型小鼠15~35 min的舔后足累计时间(P〈0.05),能明显抑制二甲苯致小鼠耳肿胀、醋酸致小鼠腹腔毛细血管通透性增高、棉球致小鼠肉芽组织增生以及角叉菜胶致气囊炎症模型小鼠的炎症反应(P〈0.01或P〈0.05);并能降低气囊炎症模型小鼠炎性渗出液中NO、TNF-α水平,减少血清中MDA水平,提高SOD活力(P〈0.01或P〈0.05)。结论地奥心血康的抗炎作用可能与抑制炎性因子NO、TNF-α及抗氧化有关。 AIM To investigate the anti-inflammatory and analgesic effects of Diao Xinxuekang(Dioscorea Panthaica Prain et Burkill,Dioscorea nipponica Makino)(DAXXK). METHODS The anti-inflammatory and analgesic effects were performed on the mice models for pain induced by formalin,of the ear swelling induced by xylene,for the capillary hyperpermeability in abdominal cavity induced by acetic acid,for the granuloma induced with cotton pellet,and for the air-pouch acute inflammation induced by carrageenan. The anti-inflammatory mechanisms were evaluated by measuring the levels of tumor necrosis factor(TNF-α) and nitric oxide(NO) in synovitis exudates determined by ELISA and nitrate reductase. Levels of SOD and MDA in serum were determined by watersoluble tetrazolium salt(WST-1) and thiobarbituric acid(TBA) methods. RESULTS DAXXK(80,60 mg/kg) could reduce the cumulative time for licking hind at intervals of 15- 35 min(P〈0. 05); lessen the xylene-induced ear edema,lower the capillary permeability in abdominal cavity and inhibit granuloma,decrease the total protein content and the amount of WBC in exudates(P〈0. 01 or P〈0. 05),reduce TNF-α and NO levels in exudates and MDA contents in serum,and increase SOD activity(P〈0. 01 or P〈0. 05). CONCLUSION DAXXK has remarkable anti-inflammatory and analgesic effects,which may be related to the inhibition of inflammatory cytokine NO and TNF-α,and antioxidant function.
