摘要
目的探讨天冬总皂苷含量的测定方法,优化超声提取工艺。方法以菝葜皂苷元为对照品,采用紫外分光光度法测定总皂苷的含量,用单因素实验和正交实验优化超声提取工艺。结果菝葜皂苷元质量浓度在5.2~52μg/m L之间线性关系良好(r=0.9994),总皂苷平均回收率达到98.83%(RSD=1.88%)。最佳超声提取工艺为乙醇浓度50%、料液比1:20(g/m L)、提取温度60℃、提取时间20 min。结论总皂苷含量测定方法简单、易行、准确,超声提取工艺高效、快捷,易于推广。
Objective: To develop a method for determining the content of total saponins in Asparagus cochinchinensis and optimize its ultrasonic extraction process. Methods:Sarsasapogenin as the control sample, the content of total saponins was determined by ultraviolet spectrophotometry. Optimization of the ultrasonic extraction process of total saponins was performed using single factor and orthogonal array design methods. Results:The calibration curve of the sarsasapogenin was linear (r=0.9994) in the range of 5.2~52μg/mL with average spike recovery of 98.83%(RSD=1.88%). The optimum ultrasonic extraction conditions were found to be:ethanol concentration of 50%, solid-to-liquid ratio of 1:20, extraction temperature of 60℃, ultrasonic extraction duration of 20 minutes. Conclusion:The developed method for total saponins determination is simple, feasible and accurate. The optimized extraction process is efficient and quick, and easy to popularize.
