摘要 目的应用随机扩增多态性DNA(RAPD)技术对肺癌患者感染白假丝酵母菌和热带假丝酵母菌进行基因分型,并分析不同基因型对不同抗真菌药物的耐药情况。方法采用经优化的随机扩增多态性DNA(RAPD)方法,对36株白假丝酵母菌和8株热带假丝酵母菌进行基因多态性分型,并对分型菌株进行药敏试验。结果 36株白假丝酵母菌可分为:13株为A型,10株为B型,7株为C型,6株为D型;8株热带假丝酵母菌可分为:5株为A型,2株为B型,1株为C型。药敏结果显示,白假丝酵母菌B型对6种抗真菌药物的敏感率明显高于A、C、D型;C型对6种抗真菌药物的耐药率明显高于A、B、D型。热带假丝酵母菌C型对6种抗真菌药物的敏感率为100%,热带假丝酵母菌A、B对伊曲康唑均出现了耐药,对其他5种抗真菌药物敏感。结论 RAPD技术用于念珠菌种内的分型具有较高的灵敏性与特异性,不同型别的白假丝酵母菌和热带假丝酵母菌耐药性可能与特定基因型有一定相关性。 Objective To genotype Candida albicans and Candida tropicalis in lung cancer patients using randomamplified polymorphic DNA(RAPD) technology and analyze drug resistance of genotypes to different antifungal drugs.Methods The optimization of random amplified polymorphic DNA(RAPD) method, 36 strains of Candida albicans and 8strains of Candida tropicalis were polymorphism, and the type strains were drug sensitive test. Results The 36 Candidaalbicans strains were divided into13 genotype A, 10 gneotype B, 7 genotype C, 6 genotype D. Eight Candida tropicalis strainswere divided into 5 genotype A, 2 genotype B, 1 genotype C. The drug sensitivity results showed, the susceptibility of Candidaalbicans type B to 6 antifungal drugs was significant higher than the types A, C and D;The sensitivity of type C to 6 antifungaldrugs was significantly higher than that of types A, B and D. The susceptibility of Candida tropicalis type C to 6 antifungaldrugs was 100%, the Candida tropical is types A and B was resistant to itraconazole while it was still susceptible to other fiveantifungal drugs. Conclusion The specificity and sensitivity of RAPD technology is high in genotyping Candida specie andthe drug resistance of different types of Candida albicans and Candida tropicalis might be associated with specific genotypes.
