两种分离大鼠触须部毛乳头改良方法比较

摘要 目的探索简便快捷分离大鼠触须部毛乳头的方法,为进一步研究毛囊再生提供细胞模型。方法分别用改良两步酶法和改良显微解剖法分离培养7日龄SD大鼠触须部毛乳头,并比较获得的毛乳头个数、形态、贴壁率、细胞迁出时间等。结果改良两步酶法总操作时间(6.91±0.37)h,较改良显微解剖法(5.66±0.40)h长(P<0.05),获得的总毛乳头数量前者(493.33±25.03)个,较后者(41.66±7.52)个多(P<0.05),其中触须型毛乳头数量前者(95.00±10.48)个,较后者(42.0±6.89)个多(P<0.05);改良两步酶法的人工操作时间(1.91±0.37)h,较改良显微解剖法(4.75±0.52)h短(P<0.05);而在贴壁时间、细胞迁出时间上两种方法无统计学差异(P>0.05);改良两步酶法获得的毛乳头间杂质细胞较多,而改良显微解剖法获得的毛乳头形态较好,纯度较高。结论改良显微解剖法适用于快捷提取较纯的触须型毛乳头,而改良两步酶法适用于简便大批量的提取触须部毛乳头。
