肾下腹主动脉移植骨髓间充质干细胞治疗脊髓缺血再灌注损伤大鼠模型的建立 被引量:4

摘要 干细胞移植是治疗脊髓损伤的研究热点和重要方向,目前移植途径多为局部注射和静脉注射.本研究在建立大鼠脊髓缺血再灌注损伤(SCIRI)改良模型基础上,经肾下腹主动脉向缺血节段脊髓分别输注不同数量Hochest 33342标记的骨髓间充质干细胞(BMSCs)悬液.采用BBB评分观察大鼠后肢功能变化,镜下观察BMSCs体内分布情况.结果显示,移植100万和200万细胞数组L2脊髓内荧光细胞分布率和术后6d、12d BBB评分显著高于50万细胞数组和培养基输注组.以上结果说明经肾下腹主动脉输注的BMSCs能够在缺血再灌注损伤脊髓内存活并促进脊髓功能恢复,最佳移植细胞数为每只大鼠推注100万细胞. Stem cell transplantation has been a hot and important direction in therapy of spinal cord injury.The most popular ways for transplantation are local injection and intravenous injection.In our study,an improved model on spinal cord ischemic reperfusion injury(SCIRI)in rats is established.Different number of bone marrow mesenchymal stem cells (BMSCs)which are labeled by Hochest 33342 are transplanted to the injured cord through the abdomen aorta respectively.The neurological functional status of rats is assessed by BBB scores at 1 , 6 and 1 2 days after the operation.After last scoring,the lumbar segments of spinal cord (L2 )are removed for observing the distribution of the BMSCs in vivo.Results show that the distribution of fluorescence cells and the BBB scores in the groups of 1×1 06 and 2 ×1 06 transplanted cells are much higher than those in the groups of 0.5 ×1 06 transplanted cells and culture medium (P〈0.01 ).These data suggest that BMSCs transplanted through the abdomen aorta can survive in the ischemia reperfusion injured cord and improve its functional rehabilitation.The desirable number of transplanted BMSCs is 1 ×1 06 cells for one rat.
