DNA去甲基化酶TET2在胃癌细胞低表达与肿瘤转移的相关性 被引量:1

摘要 目的探讨DNA去甲基化酶TET2和E-钙黏蛋白(E-cad)在胃癌组织的表达及临床意义。方法收集该院行手术切除的胃癌标本100例,采用免疫组化技术检测胃癌组织中TET2和E-cad的表达,并分析其与患者临床病理特征及预后的关系。结果 TET2和E-cad在胃癌组织高表达率分别为24%(24/100),26%(26/100),显著低于癌旁黏膜82.8%(21/29),65.5%(19/29)(P<0.05)。TET2和E-cad在TNMⅢ期、发生淋巴结转移的胃癌组织表达率明显降低(P<0.05)。TET2或E-cad低表达组患者总生存率和无转移生存率明显低于高表达组(P<0.05)。胃癌组织中TET2与E-cad表达呈显著正相关(P<0.01)。结论 TET2与E-cad表达呈正相关,胃癌细胞TET2低表达与胃癌转移有密切关系,提示预后不良。 Objective To investigate the expression of DNA demethylation enzyme TET2 and E-cadherin in gastric cancer tissue and their clinical significance. Methods The expressions of TET2 and E-cadherin protein in gastric cancer tissue specimens from 100 operatively treated patients in our hospital were examined by immunohistochemieal staining. Their correlation with several clinicopathological characteristics and prognosis was anlyzed. Results The high expression rates of TET2 and E-cadherin in gastric cancer tissue were 24% (24/100) and 26 % (26/100) respectively, which were significantly lower than 82.8% (21/29) and 65.5 % (19/29)in the adjacent normal mucosa(P〈0.05). The expression rates of TET2 and E-cadherin in the gastric caner tissue with TNM stage Ⅲ and limph node metastasis were significantly decreased (P〈0. 05). The total survival rate and the non--metastasis survial rate in the TET2 or E-cadherin low eapression group was significantly lower than those in the TET2 or E-cadherin high eapression group(P^0.05). The TET2 expression was positively correlated with the expressions of E-cadherin in the gastric cancer tissue(P^0.01). Conclusion The TET2 expression is positively correIated with the expressions of E-cadherin. The low expressin of TET2 is closely related with gastric cancer metastasis,which indicates the poor prognosis.
