摘要
以苎麻品种"赣苎三号"种子萌发的无菌苗的下胚轴为外植体,通过研究不同培养基、不同生长调节物质对苎麻种子萌发、下胚轴愈伤诱导、不定芽的发生和生根的影响,筛选出以"赣苎3号"下胚轴为外植体的组织培养再生体系。结果表明:1/2 MS+TDZ 0.5 mg/L+IAA 0.01 mg/L+2,4-D 0.03 mg/L+CH 500 mg/L+蔗糖30 g/L+琼脂7 g/L为最佳种子萌发培养基,种子萌发率为77%。1/2 MS+TDZ 0.3 mg/L+IAA 0.03 mg/L+2,4-D 0.02 mg/L+CH 300 mg/L+蔗糖30 g/L+琼脂7 g/L为愈伤组织诱导及分化最佳培养基,愈伤率为98%,出芽率为51%。1/2 MS+蔗糖15 g/L+琼脂7.5 g/L为愈伤苗的最佳生根培养基,生根率为100%。
A tissue culture regeneration protocol had been established for plantlet regeneration from hypocotyls induced callus of "Ganzhu 3" Boehmeria nirea, by studying the effects of different culture medium and plant hormone on seed germination, callus inducement of hypocotyls, adventitious bud regeneration and rooting of B. nirea. Results showed that: The optimum medium for seed germination is 1/2 MS+TDZ 0.5 mg/L+IAA 0.01 mg/L+2,4-D 0.03 mg/L+CH 500 mg/L+ Sucrose 30 g/L+Agar 7 g/L, and germination rate could reach the maximum of 77%. The optimum medium for the callus inducement and differentiation is 1/2 MS + TDZ 0.3 mg/L + IAA 0.03 mg/L + 2, 4-D 0.02 mg/L + CH 300 mg/L + Sucrose 30 g/L +Agar 7 g/L, with 98%callus inducing rate and 51%differentiation rate. The best rooting medium is 1/2 MS+Sucrose 15 g/L+Agar 7.5 g/L, with 100%rooting rate.
