摘要
目的:探讨虎杖苷治疗非酒精性脂肪肝的疗效。方法:(1)造模:健康雄性SD大鼠47只,随机分为5组:正常组(10只)、模型组(10只)、虎杖苷大剂量组(9只)、虎杖苷小剂量组(9只)和双环醇组(9只)。正常组予标准饲料,其它4组给予高脂饲料;5组共喂养10周后,从正常组与模型组中各随机挑选1只,取肝脏做病理切片,正常组大鼠肝脏切片作对照,证实模型组大鼠非酒精性脂肪肝的形成。(2)治疗:正常组与模型组予以生理盐水灌胃,5 m L/次;虎杖苷大、小剂量组分别以200 mg·kg-1·d-1,100 mg·kg-1·d-1的虎杖苷混悬液灌胃,双环醇组以100 mg·kg-1·d-1的双环醇混悬液灌胃。以上5组皆观察4周,期间各组大鼠统一用标准饲料饲养。次日大鼠禁食12 h后,腹腔麻醉,1抽取腹主动脉血5 m L,分离血清,检测大鼠血清中肝功(丙氨酸氨基转移酶ALT、天门冬氨酸氨基转移酶AST)、血脂(总胆固醇TC、甘油三酯TG);2取肝组织,在大鼠肝脏的同一部位取4块黄豆大小肝组织:一块投多聚甲醛浸泡,为HE染色观察肝脏病理学改变准备。另两块装Ep管,速投液氮。一份观察油红O染色观察肝细胞内脂质沉积状况。结果:1大鼠体质量变化:造模第10周末,模型组、虎杖苷两个剂量组和双环醇组的大鼠体质量明显高于正常组(P<0.05,0.01);治疗1周后,与模型组相比虎杖苷大剂量组和双环醇组体质量开始出现降低(P<0.05,0.01),治疗第4周末,正常组、虎杖苷两个剂量组及双环醇组大鼠体质量低于与模型组,差异具有统计学意义(分别对应为P<0.05,0.01),且虎杖苷大剂量对大鼠体质量减轻效果与双环醇组相当(P<0.05)。2肝功血脂的变化:治疗4周后,与模型组相比,TC、TG在虎杖苷两个剂量组、双环醇组中均得到明显改善(P<0.05,0.01)。虎杖苷大剂量组对TG的降低作用优于双环醇组(P<0.05)。3肝脏病理的变化:治疗4周后,与模型组相比,肝细胞脂肪变、气球样变、�
Objective : To investigate the effects of polydatin on nonalcoholic fatty liver. Methods : 1 model : 47 healthy male SD rats, were randomly divided into 5 groups: normal group (10 rats) , model group (10 rats), polydatin in high dose group (9 rats) , polydatin in small dose group (9 rats) and bicyelol group (9 rats). Normal group received standard diet, the other 4 groups were fed with high -fat diet, a total of 5 groups after 10 weeks of feeding, the normal group and model group randomly 1 only, the liver pathological slices, the normal group rats liver sections as controls, confirmed the formation of alcoholic fatty liver of rats in model group than in the. 2 treatment : normal group and model group were given normal saline ,5 mL/times; polydatin was high, low dose group were treated respectively with polydatin 200 mg ~ kg-1 . d-I ,100 mg ~ kg-1 ~ D-1 suspension gavage, bicyclol group with bieyclol 100 mg ~ kg-l ~ D-l suspension at the stom- ach. The above 5 aTOUnS were observed for 4 weeks_ durin~ each standard feed rats unitv. The rats were fasted for 12 hours after the extraction, anesthesia, abdominal aortic blood 5ml, serum separation, detection of liver in rat serum (ala- nine aminotransferase ALT, aspartate aminotransferase (AST) , serum total cholesterol TC, glycerin three greases TG); liver tissue, four pieces. The size of liver tissue in the same position of rat liver: a cast of paraformaldehyde immer- sion, for HE staining to observe the pathological changes in the liver. The other two Ep pipe, speed into liquid nitrogen. Observe the deposition of lipid in the liver cells were observed by oil red O staining. Results : the changes of body weight of rats: the model tenth weekend, polydatin from model group, two dose groups and bicyclol group the body weight of rats was significantly higher than that in normal group ( P 〈 0.05, O.
