摘要
唾液是蚜虫与寄主植物相互作用的关键因子,蚜虫唾液研究对于作物抗蚜品种选育、防治技术开发和虫瘿形成机理研究等具有重要意义,但唾液的提取和浓缩是研究中的难点。本研究根据角倍蚜体型微小、体表有蜡粉的特点,对蚜虫唾液研究中常用的双层Parafilm膜夹营养液方法进行了改进,改进的方法能够成功提取角倍蚜干雌的唾液,用于质谱分析和蛋白鉴定。
Aphid saliva plays a crucial role in aphid- host plant interactions. It has important significance in breeding for aphid resistant variety crops,development of pest management techniques,and exploration for gall formation mechanism. However,it is so difficult for saliva research that just several species have been collected and analyzed in saliva during the past fifty years. The paper introduces an improvement method based on the current aphid saliva collection method,a double layer membrane of Parafilm with an artificial diet which has been used for free living aphids with large size body. We have collected and analyzed saliva successfully from Schlechtendalia chinensis,a galling aphid with tiny size body,using the improvement method. It may be used for saliva research of other aphids with tiny size body.
