摘要
目的阐述miR-1976及CD105,整合素αvβ6的关系,探讨三者在阴道感染中的作用及机制。方法采用Real-Time PCR方法检测健康及细菌性阴道病患者阴道黏膜组织中miR-1976与CD105、整合素αvβ6的表达;评估阴道上皮细胞系VK2/E6E7过表达miR-1976后其CD105,整合素αvβ6的表达情况;阐述过表达miR-1976对VK2/E6E7细胞增殖和凋亡的影响。结果细菌性阴道病患者阴道黏膜组织中miR-1976显著低表达,而CD105,整合素αvβ6的mRNA显著高表达(P<0.05);VK2/E6E7细胞过表达miR-1976后,其增殖和凋亡未受明显影响,而CD105和整合素αvβ6的mRNA则显著低表达(P<0.05)。结论 miR-1976通过CD105调节整合素αvβ6的表达,从而影响阴道上皮细胞的黏附能力,参与细菌性阴道病的发生。
Objective To clarify the relationship among miR-1976,CD105 and integrinαvβ6 and to evaluate the function and mechanism of miR-1976,CD105 and integrin αvβ6 in vaginal mucosa infection. Methods The expression levels of miR-1976,CD105 and integrin αvβ6 in vaginal mucosa of healthy women and vaginitis ones were detected by real-time PCR,and the expression levels of CD105 and integrin αvβ6 in VK2 / E6E7 cell lines were investigated under the circumstance of miR-1976 over-expressed. Meanwhile,flow cytometry was used to evaluate the influence of lentivirus on cell proliferation and apoptosis. Results The expression of miR-1976 was significantly lower in vaginitis mucosa than in healthy ones( P〈0. 05),and the mRNA of CD105 and integrin αvβ6 were much higher reversely( P〈0. 05). After miR-1976 was over-expressed in VK2 / E6E7 cell lines,the cell proliferation and apoptosis were scarcely impacted by the lentivirus,and the mRNA of CD105 and integrin αvβ6 were much lower than before( P〈0. 05). Conclusion It has been verified that miR-1976 can regulate the expression of integrin αvβ6 through CD105,and miR-1976 also participate in vaginal mucosa infection through influencing the adhesion ability of vaginal mucosa cells.
