摘要 目的:对华支睾吸虫硫氧还蛋白跨膜蛋白(CsTMX)基因进行生物信息学分析。方法利用生物信息学方法(InterProScan、SignalP、TMHMM和SWISS—MODEL等相关软件)对CsTMX基因及相应氨基酸序列的同源性、理化性质、保守结构域、信号肽、亲水性/疏水性、三级结构进行预测分析。结果 CsTMX基因的开放阅读框(ORF)包含414 bp,编码137个氨基酸,理论分子质量为16.04±103,等电点为5.16。TMHMM和SignalP3.0分析结果显示CsTMX含信号肽,其在第19~20位氨基酸之间有1个切割位点;InterProScan和SWISS—MODEL分析结果显示,TMX特征性结构域位于第21~27位氨基酸之间,并且具有高度保守的CPAC基因序列。结论利用生物信息学知识和各种分析软件对CsTMX所编码的cDNA序列及理论蛋白质的理化性质、结构和功能域等进行预测和分析,能够为开展CsTMX的表达及其功能研究提供理论依据。 Objective To analyze protein characteristics of thioredoxin—related transmembrane protein gene of clonorchis sinensis(CsTMX) by bioinformation. Methods The homology,physicochemical property,conserved domain,signal peptide,hy—drophilcity,hydrophobicity and the tertiary structure of CsTMX and amino acid sequence were predicted by bioinformatic analysis software such as InterProScan,SignalP,TMHMM and SWISS—MODEL. Results The open reading frame of CsTMX cDNA in—cluded 414 bp,coded 137 amino acids with theoretical molecular weight being16.04 ±103 and isoelectric point being 5.16. TMH—MM and SignalP3.0 analysis showed that CsTMX contained signal peptide,having a cleavage site between 19th and 20th aminoacids. InterproScan and SWISS—MODEL analysis indicated the characterized TMX domain was found in the position of 21st-27th amino acids being possess with highly conserved CPAC gene sequence. Conclusion The bioinformatics analysis and vari—ous software may predict and analyze cDNA sequences coded by CsTMX and physicochemical property,structure and function do—main of theoretical protein,which is available for providing theoretical basis of expression of CsTMX.
