摘要
目的研究蒙药复方森登-4全方有效部位对佐剂型关节炎(AA)模型治疗效果,为其进一步开发利用奠定基础。方法 SD大鼠随机分成蒙药复方森登-4有效部位组、雷公藤组、塞来昔布组、模型组、正常组。除正常组以外,所有组采用完全弗氏佐剂导致关节炎模型,二次免疫以后给药;用自制足爪容积测量器测定每只大鼠的双足体积,计算大鼠足爪的肿胀度;用ELISA试剂盒检测血清中肿瘤坏死因子-α(TNF-α)、前列腺素-2(PGE2)及白细胞介素-6(IL-6)炎性因子的水平。结果给药20 d后,蒙药复方森登-4有效部位组、塞来昔布组大鼠足肿胀度与模型组比较有非常显著的差异(P<0.01),而雷公藤组大鼠的足肿胀度与模型组比较有差异(P<0.05);这3组治疗组与正常大鼠的足肿胀度没有显著性差异。在检测TNF-α、PGE2、IL-6的水平时,雷公藤组、塞来昔布组与模型组相比有差异(P<0.05),而蒙药复方森登-4有效部位组与模型组比较差异显著(P<0.01)。结论蒙药复方森登-4对类风湿性关节炎不同的炎症因子有很强的抑制作用,而类风湿性关节炎的炎症因子降低与类风湿性关节炎的有效治疗正相关,所以蒙药复方森登-4可以有效治疗类风湿性关节炎。
Objective To study the therapeutic effect of effective part of Sendeng- 4, a Mongolian medicine compound, on rat model with adjuvant arthritis (AA). Methods SD rats were randomly divided into five groups: effective part of Sendeng- 4 group (Sendeng- 4 group) , Tripterygium wilfordii Hook F. group (TWHF group) , celecoxib group, model group and normal group. Except the normal group, the other groups were made into AA model induced by injection complete Freund' s adjuvant into both rear voix pedis twice and were orally administrated corresponding drugs for 20 consecutive days after the second immunization. The rat paw swelling degree were calculated by measurement of the volume of rat paw. The concentration of serum tumor necrosis factor-α (TNF-ct), prostaglandin-2 (PGE:) and interleukin-6 (IL-6) were detected by using ELISA kit. Results Compared with model group, the rat paw swelling degree decreased significantly in Sendeng- 4 group, TWHF group and celecoxib group (P 〈 0.05 ), while, those of the three treatment groups had no difference compared with normal group. Compared with model group, the concentration of TNF-ct, PGE2 and IL-6 decreased significantly in three treatment groups ( P 〈 0.05 ) . Conclusion Mongolian medicine Sendeng- 4 inhibites TNF-a, PGE2 and IL-6 in serum which are regarded as rheumatoid arthritis inflammatory cytokines, leading to effective treatment of the patients of rheumtoid arthritis.
