摘要
目的:探讨蝙蝠葛碱(DAU)对阿尔茨海默病(AD)小鼠学习记忆及海马白介素-12(IL-12)的影响。方法:小鼠随机分成对照组、模型组、吡拉西坦组(0.62 g/kg/d)、DAU高剂量组(1.17 g/kg/d)、DAU低剂量组(0.59 g/kg/d)。采用Aβ_(1-42)双侧脑室注射小鼠诱导AD模型。给予DAU灌胃,治疗18d后取材。应用Morris水迷宫检测小鼠行为学指标及双抗体夹心法检测海马IL-12的水平。结果:与模型组比较,DAU能改善AD模型小鼠的学习和记忆(P<0.05),降低海马IL-12水平(P<0.05)。结论:DAU能改善AD模型小鼠学习记忆功能,DAU通过抑制IL-12在AD治疗中发挥重要作用。
Objective: To observe the effects of Dauricine(DAU) on learning and memory and Intedeukin-12(IL-12) of Hippocampal in Alzheimer's disease (AD) mouse. Methods: Mice were randomly divided int: control group, model group, Piraeetam group(0.62 g/kg/d), DAU high-dose group(1.17g/kg/d), DAU low-dose group(0.59g/kg/d). Aβ-42 was injected into the bilateral hippocampus of mouse to induce an AD model. The mice were killed after 18 days continuous treating. After the treatment all animals were sacrificed. The Morris water maze was used to observe the learning and memory for the mouse. Biochemical methods were used to determice the content of IL-12 in the Hippocampal. Results: Compared with the model group, DAU could significantly improve the learning and memory of AD model mouse(P〈0.05). DAU could decrease the eonten of IL-12(P〈 0.05).Conclusion: DAU could improve the ability of learning and memory of AD model mouse. DAU plays a certain role in the treatment of AD through inhibiting IL-12.
