转基因水稻多重PCR和Real-time PCR定性检测中的问题与策略

摘要 随着转基因水稻研究在世界范围内的研究和开发,以及转基因水稻商业化的争议性加剧,相关部门对粮食安全、食品安全更加关注,农业科研单位检测转基因水稻也将具有重要意义。参照农业部的质检标准书和转基因PCR检测中的实践经验,对抗虫转基因水稻在转基因检测中可能出现的问题进行了技术层面和理论层面分析;同时对转基因抗虫水稻的启动子、终止子、内参基因、靶基因等几个元件的检测分别进行探讨,并重点分析了启动子和终止子的检测工作,根据研究者的工作经验报道,从植物病毒学原理和植物基因工程的理论角度,提出了对转基因PCR定性检测中可能遇到问题的解决方案。 As genetically-modified(GM) rice will be commercialized, qualitatively detecting GM rice becomes more and more important. Based authors' practice in detecting of transgenic Bt rice with the quality standard of Chinese Ministry of Agriculture, the potential problems were analyzed from technical level and theoretical perspective. The detection of transgenic components(promoter, terminator, reference gene and target gene etc) of GM rice with PCR was discussed. The possible solutions were proposed from the principles of plant virology and genetic engineering based on previous reports.
