马齿苋高频再生体系的建立及对草丁膦敏感性测定 被引量:2

Establishment of High Frequency Regeneration System of Portulace oleracea L.and Determination of Phosphinothricin Sensitivity

摘要 以马齿苋子叶和下胚轴为外植体,研究了外源激素对其不定芽的诱导及不同浓度6-BA、NAA、GA3对不定芽伸长的影响,并测定了马齿苋子叶和下胚轴对草丁膦的敏感性。结果表明:MS+2.0mg/L 6-BA+100mg/L水解酪蛋白为下胚轴不定芽再生最适培养基,不定芽再生率达96.65%;MS+3.0mg/L 6-BA+100mg/L水解酪蛋白为子叶不定芽再生最适培养基,不定芽再生率达100%;最佳的不定芽继代培养的培养基为MS+0.5mg/L 6-BA+0.05mg/L NAA+0.2mg/L GA3;生根培养基为MS+2.0mg/L IBA,生根率达100%;以子叶和下胚轴为材料研究其对草丁膦的敏感性,发现子叶对草丁膦更敏感,0.050mg/L草丁膦对子叶的致死率达到89.0%;对下胚轴的致死率为71.5%。该研究建立了马齿苋植株高频再生体系,并确定了其对草丁膦的敏感性,可为该品种的基因工程研究提供必要的理论和实践基础。 Taking Portulaca oleracea L.cotyledon and hypocotyl as explants,the effect of exogenous hormone on the inducement of adventitious buds,and different concentration of 6-BA,NAA,GA3 on the elongation of adventitious buds were studied,the sensitivity of phosphinothricin of Portulaca oleracea L.cotyledon and hypocotyl were determined.The results showed that the optimum induction mediun of hypocotyl adventitious buds was MS+2.0mg/L 6-BA+100mg/L CH(casein acid hydrolysate)and the average induction rate was 96.65%;the optimum mediun of cotyledon adventitious buds induction was MS+3.0mg/L 6-BA+100mg/L CH(casein acid hydrolysate)and the average induction rate was100%;the optimum shoot growth medium was MS+0.5mg/L 6-BA+0.05mg/L NAA+0.2mg/L GA3;the rooting medium was MS+2.0mg/L IBA with rooting rate 100%.Determination of the phosphinothricin sensitivity showed that compared to hypocotyl,cotyledon was more sensitive to phosphinothricin.At 0.050 mg/L PPT,the fatality rate was89.0%and 71.5% in cotyledon and hypocotyl respectively.This study established the high frequency regeneration system of Portulace oleracea L.and determined its sensitivity to phosphinothricin.Results from this study would help the research of Portulaca oleracea L.in genetic engineering.
