摘要
目的:探讨温经汤对子宫内膜异位症大鼠在位和异位内膜中血管内皮生长因子(VEGF)及富含半胱氨酸的酸性分泌蛋白(SPARC)表达的影响。方法:自体移植法建立子宫内膜异位症(EMs)大鼠模型。将造模成功的大鼠随机分为模型组、米非司酮组及温经汤低、中、高剂量组,另取一组正常大鼠为正常对照组。各组分别给予生理盐水、米非司酮和温经汤灌胃,连续给药21 d后处死大鼠,取子宫和异位内膜组织。免疫组化法观察在位和异位内膜中VEGF和SPARC的定位和表达情况,Western blotting法检测二者在在位和异位内膜中的蛋白表达量。结果:免疫组化结果显示,模型组在位和异位内膜中VEGF和SPARC阳性表达强烈,各治疗组在位和异位内膜中二者的阳性表达均有不同程度的降低。Western blotting检测显示,温经汤中、高剂量组在位和异位内膜VEGF蛋白含量较模型组显著降低(P<0.05);除温经汤低剂量组外,其余各组在位内膜SPARC蛋白含量均低于模型组(P<0.05);温经汤中、高剂量组异位内膜SPARC蛋白含量和模型组比较,差异有统计学意义(P<0.05)。结论:温经汤可以降低EMs大鼠在位和异位内膜中VEGF和SPARC表达,影响新生血管形成,抑制异位子宫内膜的生长。
Objective:To explore the effect of "Wenjing Decoction" on the expressions of VEGF and SPARC in eutopic and ectopic endometrial tissues of endometriosis rats. Methods : The models of endometriosis was established by transplantation autoplastic and randomly divided into 5 groups: model group, Mifepristone group and "Wenjing Decoction" groups (low, medium and high dose), while another group of rats were divided into the normal control group. Rats were respectively treated with normal saline, mifepristone and " Wenjing Decoction", After 21-day treatment, the rats was sacrificed. The tissues of uterus and eutopic and ectopic endometrial were taken out. The expression localization of VEGF and SPARC in eutopic and ectopic endometrial tissues were detected by immunohistochemical method, and their protein quantities were assessed by Western blotting. Results: Results of immunohistochemical method showed that the positive expression of VEGF and SPARC in eutopic and ectopic endometrium were strong in the model group, which was decreased in different degrees in each treatment groups. Results of Western blotting showed that compared with model group, the protein expression of VEGF in medium- and high-dose "Wenjing Decoction" groups was obviously decreased (P 〈 0.05 ). Except for low-dose "Wenjing Decoction" group, the protein expression of SPARC in eutopic endometrium was lower than that of the model group ( P 〈 0.05 ). There was a significant difference in the protein expression of SPARC in ectopic endometrium in medium- and high- dose "Wenjing Decoction" groups compared with the model group ( P 〈 0. 05 ). Conclusion : " Wenjing Decoction" can decrease the expressions of VEGF and SPARC in eutopie and ectopic endometrial tissue in endometfiosis rats, influence the angiogenesis, and inhibit the growth of ectopic endometrium.
