摘要
用不同浓度的胰岛素培养人肾小管上皮细胞株48h后,采用实时定量PCR检测人尿酸盐转运子mRNA的相对表达量。随胰岛素浓度的增加,人尿酸盐转运子mRNA表达水平明显降低(5、25、125、500μIU/ml时的相对表达量分别为0.95、0.40、0.24、0.23)。提示离体培养条件下的肾小管上皮细胞人尿酸盐转运子基因表达改变与胰岛素浓度的变化有关。
Human proximal tubule epithelial cell line, HK-2 cells, were cultured with various concentrations of insulin for 48 h. Human urate transporter (hUAT) mRNA was detected by reahime quantitative PCR. hUAT mRNA levels were down-regulated by insulin (5,25,125,500 μIU/ml) in a dose-dependent manner ( relative expression median were 0.95, 0.40, 0.24, and 0.23 ). In vitro, the expression of hUAT mRNA in HK-2 cells is associated with the concentration of insulin.
