参麦合剂对兔干眼眼表MUC5AC表达的影响 被引量:1

摘要 目的观察参麦合剂对兔干眼模型眼表黏蛋白5AC(MUC5AC)表达的影响。方法 40只健康新西兰大白兔随机分为空白对照组、造模组、生理盐水组、中药治疗组,后3组采用0.1%苯扎氯胺滴眼液建立干眼模型。生理盐水组和中药治疗组分别给予等量生理盐水和中药灌胃,灌胃给药第7、14、21、28天同期收集各组泪液,ELISA法检测MUC5AC含量。给药第28天,每组随机选取5只兔,取同一部位的结膜组织行Westen blot和免疫荧光检测MUC5AC的表达。结果完成造模后,造模组、生理盐水组和中药治疗组的泪液MUC5AC含量均明显低于空白对照组,差异有统计学意义(P<0.01)。中药治疗组泪液MUC5AC含量随给药时间延长逐渐升高,至给药28 d时与空白对照组数值接近(P>0.05),其结膜组织中MUC5AC的表达也高于造模组和生理盐水组(P<0.05),与空白对照组相近(P>0.05)。结论参麦合剂能增加兔干眼模型MUC5AC在泪液及结膜的表达。 OBJECTIVE To explore the effect of Shenmai-mixture on expression of the ocular surface mucin MUC5AC in rabbit dry eye model. METHODS Forty healthy New Zealand white rabbits were divided randomly into 4 groups: the control group, dry eye model group, saline solution group and traditional Chinese medicine treated group, and 0.1% Benzalkonium chloride (BAC) was used to establish the rabbit dry eye model. Saline solution group and drug treatment group were given the same amount of saline and traditional Chinese medicine by intragastric ad- ministration. The tears of 4 groups were collected on day 7, 14, 21 and 28. ELISA was performed to detect MUC5AC content. On the 28th day, each group randomly selected 5 rabbits, took the same part of the conjunctival tissue. Westen-blot and immunofluoreseence staining were performed to detect MUC5AC expression. RESULTS The con- tent of tear MUC5AC was significantly lowered in dry eye model groups than that in the control group (P〈0.01). The content of MUC5AC in traditional Chinese medicine treated group increased gradually with the extension of treat- ment time, and was closed to the control group on day 28 (P〉0.05). On the 28th days, the MUC5AC expression of conjunctival tissue in traditional Chinese medicine treated group was also similar to the control group (P〉0.05), higher than the other two groups (P〈0.05). CONCLUSIONS Shenmai-mixture could increase dry eye rabbit model MUC5AC expression of eonjunetiva and in tears.
