摘要
目的:观察益肺通络方对低氧性PVR大鼠无肌型肺小动脉血管重建的影响。方法:采用气管注入脂多糖联合烟熏法建立COPD大鼠模型,并叠加低氧建立PVR大鼠模型。60只Wistar大鼠随机分成空白、模型、实验高、中、低剂量5组,每组12只,空白、模型组生理盐水灌胃,实验组按高剂量(含药1.5 g/mL)、中剂量(含药1.0 g/mL)、低剂量(含药0.5 g/mL)益肺通络方煎剂灌胃。4、5、6周3个时点取材,计算3型肺小动脉百分比。结果:3型肺小动脉百分比,空白组各时点无明显改变(4周≈5周≈6周),模型组无肌型动脉呈减少趋势(4周〉5周〉6周),部分肌型动脉呈增加趋势(4周〈5周〈6周),0~4周无肌型动脉减少快,4~6周减少慢。无肌型动脉百分比,实验组各时点变化趋势均同模型组,组内分析,4周~6周比较,差异均有统计学意义(P〈0.05);各时点组间比较,均表现为模型组〈实验低剂量组〈实验中剂量组〈实验高剂量组〈空白组,各实验组与模型组比较,各实验组组间比较,差异均有统计学意义(P〈0.05)。结论:益肺通络方能抑制慢性低氧PVR大鼠肌型肺小动脉中膜增殖,且存在量效关系。
Objective: To investigate the effect of Yifei Tongluo Formula on hypoxic pulmonary vascular remodeling rats' muscularization of non muscular pulmonary arterioles. Methods: COPD rat models were established by passive cigarette smoking and intratracheal LPS instillation and superpose hypoxia to build rat models of PVR. Sixty healthy Wistar rats were randomly divided into control group( n = 12),hypoxia group( n = 12),high dose group( n = 12),medium dose group( n = 12),low dose group( n = 12). Control group and hypoxial group received intragastric normal saline and treatment groups received intragastric Yifei Tongluo Formula at high dose( 1. 5 g / mL),median dose( 1. 0 g / mL),low dose( 0. 5 g /mL). At the week 4,week 5 and week 6,the samples were taken and then we calculated the percentage of 3 different types of pulmonary arterioles. Results: In 3 types of pulmonary arterioles,control group had no significant changes at different time points( week 4≈week 5≈week 6). The hypoxia group had a decreasing trend in non muscular pulmonary arterioles( week 4 〉week 5〉 week 6) and the portion had an increasing trend in muscular pulmonary arterioles( week 4〈 week 5 〈week 6). Non muscular pulmonary arterioles decreased fast from week 0 to week 4. The decrease slowed down from week 4 to week 6. Treatment groups had the same trends in percentage of non muscular pulmonary arterioles as the hypoxia group. Within group analysis,there was significant difference( P〈0. 05) when compared week 4,week 5 and week 6. When compared different time points,all appeared to be hypoxial group 〈low dose group 〈medium dose group 〈high dose group 〈control group. Comparison between treatment groups and control group and comparison of every treatment groups showed significant difference( P〈0. 05). Conclusions: Yifei Tongluo Formula can efficiently restrain HPHand there is a dose-effect relationship.
