摘要
目的:观察大鼠心肌梗死急性期使用宁心颗粒心肌内乙酰胆碱酯酶(Ach E)的活性。方法:选取清洁级SD大鼠86只,按照既往文献方法制作大鼠心肌梗死模型,术后存活的大鼠随机分为假手术组、模型组、美托洛尔组、中药大剂量组和正常剂量组。分别相应给药30d后,从各组大鼠心脏梗死区周围区域相同位置取材,用实时定量PCR法检测心肌中Ach E mRNA的水平。结果:心肌梗死后,模型组大鼠心肌内Ach E mRNA的水平相比假手术组显著升高(P<0.01),施以药物30d后,与模型组比较,中药组的Ach E mRNA水平显著降低(P<0.01)。而美托洛尔组则无明显差异。中药大剂量、正常剂量组间无明显差异。结论:心肌梗死急性期发生恶性心律失常几率较高。检测宁心颗粒用药前后心肌中乙酰胆碱酯酶的活性变化,可以了解药物对心梗后重构的迷走神经的影响。
Objective: To observe the activity changes of Acetyl cholinesterase(Ach E) in myocardum after using Ningxia Granule in the acute phase of myocardial infarction.Method: A total of 86 male SD rats were used for the experiments.According to previous literature method,the myocardial infarction model was established.The survival rats were randomly divided into sham group,model group,metoprolol group,Ningxin high-dose group and Ningxin normal dose group.Sham group and model group were gavaged with saline while the others groups were gavaged with corresponding drug respectively for 30 days.The infarcted heart tissues were taken from the same location in each group,the level of Ach E was examined by RT-PCR method.Results: Compared with the sham group,the Ach E mRNA level was significantly increased after myocardial infarction,and showed an increasing trend as time passed(P〈0.01).Compared with the model group,the Ach E mRNA level in Ningxin Granule group was significantly decreased after given drugs for 30 days(P〈0.01).There was no significant difference in metoprolol group,TCM high dose and normal dose groups.Conclusion: The studies had shown that acute myocardial infarction was always accompanied with multiple arrhythmia.And it was necessary to explore the effect of Ningxin Granule on the remodeling vagus nerve after the acute phase of myocardial infarction by detecting the activity of acetyl cholinesterase in myocardium.
