聚乙烯醇/魔芋葡甘露聚糖复合凝胶的体外药物释放特性

摘要 目的研究聚乙烯醇/魔芋葡甘露聚糖复合凝胶的体外药物释放特性,评价复合凝胶作为新型给药系统的应用。方法以盐酸小檗碱为模型药物,采用冷冻-解冻法制备聚乙烯醇/魔芋葡甘露聚糖复合凝胶;在不同体外释放介质中,对复合凝胶的溶胀动力学、溶蚀动力学和药物释放进行测定,探讨复合凝胶的体外药物释放特性。结果复合凝胶的体外药物释放随介质pH增加而增加,随介质离子强度降低而增加,随介质中酶浓度增加而增加;复合凝胶的溶胀性能几乎不受介质pH影响,随介质离子强度降低而增加,随介质中酶浓度增加而增加;复合凝胶的溶蚀性能几乎不受介质离子强度影响,在0.1mol/L盐酸介质中溶蚀增加,随介质中酶浓度增加而增加。结论聚乙烯醇/魔芋葡甘露聚糖复合凝胶可作为pH、离子强度和酶敏感给药系统用于调控盐酸小檗碱的释放。 Objective To study in vitro drug release characteristics of polyvinyl alcohol(PVA)/konjac glucomannan(KGM)complex hydrogel and to evaluate the complex hydrogel as new drug delivey system.Methods The PVA/KGM complex hydrogel containing berberine hydrochloride as model drug was prepared by freezing and thawing method.The effects of in vitro dissolution media on drug delivery,dynamic swelling and erosion percent of the complex hydrogel were studied respectively.Results The berberine hydrochloride release rate increased with the increased of pH value,the decreased of ionic strength or the increased ofβmannase concentration of the release media.The swelling ratio of the complex hydrogel was almost constant in media with different pH value.The swelling ratio of the complex hydrogel increased with the decreased of ionic strength or the increased ofβmannase concentration of the release media.The erosion percent of the complex hydrogel was almost constant in released media with different ionic strength.The erosion percent of the complex hydrogel increased in medium of 0.1mol/L HCl or with the increased ofβmannase concentration of the release media.Conclusion The PVA/KGM complex hydrogel could used as pH sensitive,ionic strength sensitive or enzyme sensitive drug delivery system for berberine hydrochloride.
