摘要
目的探讨Slit2及其受体Robo1在宫颈鳞癌(SCC)、宫颈上皮内瘤变(CIN)及正常宫颈上皮(NCE)中的表达,及其与宫颈病变发展的关系。方法组织芯片与免疫组化技术法检测55例SCC、30例CINⅠ-Ⅱ级、30例CINⅢ级及20例NCE中Slit2、Robo1蛋白的表达;实时荧光定量聚合酶链式反应法检测20例SCC、20例CINⅢ级及20例NCE中Slit2-mRNA的表达;分析患者临床特征与Slit2表达水平的相关性。结果 Slit2、Robo1蛋白阳性表达率在NCE、CIN、SCC中逐渐降低,CINⅢ级组阳性表达率显著低于CINⅠ-Ⅱ级组(P〈0.05)。Slit2蛋白表达与宫颈癌临床分期、宫颈浸润深度、淋巴结转移相关(P〈0.05);与年龄、组织分化、肿物大小无关(P〉0.05)。NCE组Slit2-mRNA的表达水平显著高于CINⅢ组和SCC组(P〈0.05)。结论 Slit2/Robo1信号通路对促癌基因表达及功能的失调控是宫颈癌发生、发展的重要因素。
Objective To investigate the expression of Slit2 and its receptor Robo1 in cervical squamous cell carcinoma( SCC),cervical intraepithelial neoplasia( CIN) and normal cervical epithelium( NCE),and to explore the relationship between Slit2 / Robo1 signal system and cervical carcinoma generation and development. Methods The technique of tissue microarray technology and immunohistochemistry were applied to examine the expression of Slit2 and Robo1 in 55 cases of SCC,30 cases of CINⅠ-Ⅱ,30 cases of CIN Ⅲ,and 20 cases of NCE. The technique of real-time polymerase chain reaction method was used to detect the expression of Slit2- mRNA in 20 cases of SCC,20 cases of CIN Ⅲ,and 20 cases of NCE. The relation between Slit2 expression and the clinical features were analyzed. Results The positive expression rate of Slit2 and Robo1 protein in NCE,CIN and SCC were on a decline curve and had statistical significance( P〈 0. 05). And the positive rate of CIN Ⅲ was much lower than that of CINⅠ-Ⅱ( P〈 0. 05). Slit2 expression in cervical carcinoma had correlation with clinical stage,the depth of invasion and lymphnode metastasis( P〈 0. 05),but had no correlation with age,tumor differentiation and tumor volume( P〈 0. 05). Conclusion Slit2 / Robo1 signal system losing its role on the expression of oncogene is the vital factors contributing to the generation and development of carcinoma.
